The method comprises the following steps: (1) inserting a photosensitive chip at the bottom of a culture bottle; wherein the photosensitive chip comprises a CO2 / pH sensing chip and an O2 partial pressure sensing chip, and a fluorescent glue layer wrapped on the surface of the sensing chip; wherein the fluorescence intensity of a first emitted light excited by the CO2 / pH sensing the chip increases as the pH of a culture solution decreases; the fluorescence intensity of a second emitted light excited by the O2 partial pressure sensingchip increases as the O2 content in the culture solution decreases; (2) placing the bacteria and the culture solution in the culture bottle, and then introducing the growth gas and sealing; (3) adjusting the temperature of the culture bottle as needed; (4) irradiating the photosensitive chip with the light source through the culture solution; and (5) collecting the fluorescence intensity of the first emitted light and the second emitted light at different time points; and determining the change of the content of the bacteria in the culture solution according to the change of the fluorescenceintensity. The bacterial culture detection method has the advantages of simple steps, low cost, low operation difficulty, good detection effect and high accuracy.
