Problems solved by technology

The use of traditional etiological inspection methods such as microscopic examination and parasite isolation method has the disadvantages of time-consuming and laborious, and the specificity and sensitivity are not high
Immunological diagnosis method, that is, to detect Toxoplasma gondii antibody, but patients with toxoplasmosis are often accompanied by low immune function, and the antibody level often decreases, making it diff

Embodiment 1

[0048] method

[0049] 1. Establishment of Toxoplasma gondii chronic infection mouse model

[0050] Mice chronically infected with the PRU strain of Toxoplasma gondii were sacrificed by cervical dislocation, and their brains were ground into a homogenate of brain tissue with sterile PBS, and 10 ul was counted under an optical microscope, repeated 3 times, and the average value was obtained. The tissue homogenate was diluted to 30 / 100ul. Toxoplasma gondii cysts were orally infected to BALB / c mice. The mice in the experimental group were fed with 30 / 100ul brain tissue homogenate, and the mice in the control group were only fed with normal saline. They were housed in separate cages, 5 per cage, and fed and watered normally.

[0051] 2. Gene microarray analysis of brain tissues of BALB / c mice infected with Toxoplasma gondii

[0052] After 2 months of infection, three mice in each of the infection group and the control group were taken, and the brain tissues were separated for R...

Abstract

The invention provides application of lncRNA 11496 in diagnosis and treatment of toxoplasmosis and belongs to the technical field of biological medicine and molecular biology. It is found through research that NONMMUG011496 (lncRNA 11496) was significantly down-regulated in the brain tissue of mice infected with Toxoplasma gondii; meanwhile, the expression of lncRNA 11496 in brain tissues of micechronically infected with toxoplasma gondii affected the proliferation, differentiation and apoptosis of nerve cells, which was closely related to Toxoplasma gondii infection. The lncRNA 11496 is a noncoding RNA first found in brain tissues chronically infected with toxoplasma gondii. Therefore, the lncRNA 11496 can serve as an important biological target, provides a basis for diagnosis of toxoplasma encephalopathy and research and development of targeted anti-toxoplasma drugs based on the lncRNA 11496, and has a good practical application value.

Description

technical field [0001] The invention belongs to the technical field of biomedicine and molecular biology, and specifically relates to the application of lncRNA11496 in the diagnosis and treatment of toxoplasmosis. Background technique [0002] The information disclosed in this background section is only intended to increase the understanding of the general background of the present invention, and is not necessarily taken as an acknowledgment or any form of suggestion that the information constitutes the prior art already known to those skilled in the art. [0003] Toxoplasma gondii is an obligate intracellular parasite. The first human case was reported in 1908. It was discovered at the same time as animal toxoplasmosis. About 500 to 1 billion people in the world are infected with toxoplasmosis. Humans have been fighting against this disease for more than a century, but the disease is still around the world. raging. In my country, about 80,000 to 90,000 newborns are born e...
