Problems solved by technology

However, in the existing design of DNA walkers, as the walking chain walks, the track for the walking chain to walk will be gradually exhausted due to the replacement of the chain or the consumption of hydrolysis, which severely limits the walking ability of the walker. The number of steps taken, which is not conducive to the continuous operation of the walker along the track

Abstract

The disclosure belongs to the technical field of DNA walker construction, and in particular relates to a track-regenerating DNA walker and its application. This disclosure proposes a concept of a track-regenerating DNA walker and verifies it. The walker uses a detachable track consisting of roadbed and rails. Its road-based chains are functionalized on gold nanoparticles, while a large number of rail chains are free in solution and will hybridize with road-based chains to construct tracks. As the walker walks along the track, the rail chains are digested and the roadbed chains are released, and the released roadbed chains continue to combine with the rail chains in the solution to form new tracks. The constant replenishment of the track allows the machine to keep walking. This machine has great advantages in signal enrichment, so it can be used for sensitive detection of Ebola virus gene fragments in serum.

Description

technical field [0001] The disclosure belongs to the technical field of DNA walker construction, and in particular relates to a DNA walker with reproducible orbit, the walker and its application in biosensing. Background technique [0002] The information disclosed in this background section is only intended to increase the understanding of the general background of the present disclosure, and is not necessarily to be regarded as an acknowledgment or any form of suggestion that the information constitutes the prior art already known to those skilled in the art. [0003] DNA molecular machines are a class of molecular machines assembled from DNA that perform machine-like behaviors at the micro or nano scale. Benefiting from the unique properties of DNA molecules, such as strict complementary base pairing, excellent recognition ability, and high programmability, DNA molecular machines can be precisely controlled and finely manipulated in response to various stimuli. To date, ...
