[0026] The experimental analysis related to the present invention will be introduced in detail below. Of course, the applicant's research and development work on the present invention is not limited thereto.

[0027] 1. Experimental part

[0028] (1) HEK293 cells express the S protein of SARS-CoV-2 (conventional method)

[0029] ① Plasmid construction of the S protein of SARS-CoV-2;

[0030] ②Establishment of cell lines stably expressing S protein;

[0031] ③ Immunocytochemical staining was used to evaluate the transfection effect;

[0032] ④Western blotting and polyacrylamide gel electrophoresis;

[0033] ⑤ SEC-MALS analysis of S protein;

[0034] ⑥Purification of S protein: The eluate was concentrated using Vivaspin 20 (GE Healthcare), and the entire concentration process was carried out in one tube with a molecular weight cut-off of 100kDa. Finally protease inhibitors completely free of EDTA were added, aliquoted, snap frozen in liquid nitrogen, and stored at -80°C un...

Abstract

The invention discloses a preparation for blocking coronavirus infection. The preparation directly acts on an oral cavity and a pharynx, can also be atomized into aerosol, and directly enters a respiratory tract and a lung along with breath. The preparation comprises galactose and a sialic acid carbohydrate chain structure. By increasing the concentration of galactose and salivary acidified glycoprotein in the oral cavity, the pharynx, the respiratory tract and the lung, coronavirus can be combined to galactose and sialic acid carbohydrate chain structure to neutralize and inhibit recognitionof coronavirus on specific receptors on the surfaces of human cells, so that the effect of blocking coronavirus is achieved, and invasion of coronavirus to human is blocked. The active ingredients ofthe preparation are preferably rich-galactose and salivary acidified glycoprotein separated and purified from milk.

Description

technical field [0001] This application relates to a preparation for blocking coronavirus infection, especially for the novel coronavirus SARS-CoV-2. Background technique [0002] Coronavirus (Coronavirus, CoV) is a single-stranded positive-sense RNA virus with an envelope, mainly using human and other mammalian cells as hosts. Although the majority of human coronavirus infections are relatively mild, they cannot be neglected due to their high infectivity. Acute respiratory syndrome coronavirus (SARS-CoV) and Middle East respiratory syndrome coronavirus (MERS-CoV) have caused more than 10,000 cases in the past 20 years, and the fatality rate of SARS-CoV is 10%, the fatality rate of MERS-CoV is 37%. On February 11, 2020, the World Health Organization and the International Committee on Taxonomy of Viruses named the novel coronavirus "Severe Acute Respiratory Syndrome Coronavirus 2" (SARS-CoV-2). [0003] SARS-CoV-2 has the ability to spread from person to person and from pe...
