Embodiment 1

[0041] Embodiment 1: the preparation of cyclo-γ-PGA modified PNIPAM hydrogel

[0042] cyclo-γ-PGA ( figure 1 ) Modified PNIPAM hydrogel: Mix the two substances according to PNIPAM / cyclo-γ-PGA in order of 100:0, 75:25, 50:50, and 25:75, and the total concentration of the solution is 20% (w / v) , fully mixed at room temperature for 3 hours, overnight at 4°C to remove air bubbles, and gelled in a water bath at 37°C. It can be observed that the dehydration of PNIPAM hydrogels has been effectively improved when PNIPAM / cyclo-γ-PGA is 75:25 or 50:50 ( figure 2 ). Under the scanning electron microscope, it was observed that when the ratio of PNIPAM / cyclo-γ-PGA was 75:25, the hydrogel showed a typical porous structure ( image 3 ).

Embodiment 2

[0043] Embodiment 2: the preparation of the cyclo-γ-PGA modified PNIPAM hydrogel of loading EGF

[0044] Mix PNIPAM with a final concentration of 150 mg / mL and cyclo-γ-PGA 50 mg / mL, mix thoroughly at room temperature for 3 hours, and place in an environment at 4°C. Fully dissolve EGF into a 1 mg / mL solution with an appropriate amount of deionized water, mix according to the ratio of hydrogel solution / EGF solution of 100:1, place in a refrigerator at 4°C, and shake on a shaker for 30 seconds every 3 minutes. The time was 30 min, and after mixing, a 10 μg / mL cyclo-γ-PGA-modified PNIPAM hydrogel solution loaded with growth factors was obtained. Finally, the homogeneously mixed solution was placed in a water bath at 37° C. to become a gel.

Embodiment 3

[0045] Example 3: Preparation of cyclo-γ-PGA modified PNIPAM hydrogel loaded with bFGF

[0046]Mix PNIPAM with a final concentration of 150 mg / mL and cyclo-γ-PGA 50 mg / mL, mix thoroughly at room temperature for 3 hours, and place in an environment at 4°C. Fully dissolve bFGF into a 1 mg / mL solution with an appropriate amount of deionized water, mix according to the ratio of hydrogel solution / bFGF solution of 100:1, place in a refrigerator at 4°C, and shake on a shaker for 30 seconds every 3 minutes. The time was 30 min, and after mixing, a 10 μg / mL cyclo-γ-PGA-modified PNIPAM hydrogel solution loaded with growth factors was obtained. Finally, the homogeneously mixed solution was placed in a water bath at 37° C. to become a gel.

Abstract

The invention relates to the field of biological materials and tissue engineering, in particular to a cyclic gamma-polyglutamic acid (cyclo-gamma-PGA) modified poly N-isopropylacrylamide (PNIPAM) hydrogel loaded with growth factors and a preparation method thereof. The hydrogel is obtained by loading growth factors such as EGF, bFGF and PDGF on the cyclic gamma-polyglutamic acid modified poly N-isopropylacrylamide hydrogel. The hydrogel which has a three-dimensional network structure and is prepared by the preparation method has good biocompatibility, can load various growth factors such as epidermal growth factor, basic fibroblast growth factor and platelet-derived growth factor to form a drug loading system capable of promoting cell proliferation and local wound healing, and can be applied to the fields of biological medicines, medical auxiliary materials and the like.

Description

technical field [0001] The invention relates to the fields of biotechnology and pharmacy, in particular to a preparation method of a cyclic gamma-polyglutamic acid modified hydrogel loaded with growth factors. Background technique [0002] Growth factors are a class of soluble polypeptide substances that regulate the growth and proliferation of specific cells by specifically binding to cell membrane receptors. They are widely used in the field of wound healing because of their strong ability to promote cell regeneration and accelerate tissue repair. According to different wounds, the required growth factors are different. For example, epidermal growth factor (EGF) can be used for superficial wounds such as diabetic skin ulcers and bedsores; basic fibroblast growth factor (basic fibroblast growth factor, bFGF) ) can promote the proliferation of vascular endothelial and smooth muscle cells, and has a deep repair effect; platelet-derived growth factor (PDGF) can stimulate fibro...
