The method comprises the steps of separation of the immune cell NK, serum-free in-vitro amplification of the immune cell NK, analysis of a growth curve of the immune cell NK, analysis of surface antigen expression of the immune cell NK, analysis of a cycle of the immune cell NK, cryopreservation and recovery of the immune cell NK and detection of various indexes of the immune cell. The immune cell NK obtained through separation via a serum-free culture medium suspension culture system has high multiplication capacity; after the cell cryopreserved with cell cryopreservation liquid 1 is recovered, the activity of the cell in the cell cryopreservation liquid 1 is the highest and is 90% or above compared with the activity of the cell in four kinds of cell cryopreservation liquid; the detection result of a flow cytometry shows that the cell before cryopreservation and the cell after cryopreservation resuscitation are not obviously different in immunophenotype and both have the immunophenotype of immune cell; and the result of flow cytometry cell apoptosis detection experiment shows that after the cells cryopreserved in the cell cryopreservation liquid 1 are resuscitated, the number of cells undergoing advanced apoptosis is minimum, and the number of cells undergoing non-apoptosis is maximum.
