Embodiment Construction

[0037] In order to enable those skilled in the art to better understand the solutions of the present invention, the following will clearly and completely describe the technical solutions in the embodiments of the present invention in conjunction with the drawings in the embodiments of the present invention. Obviously, the described embodiments are only It is an embodiment of a part of the present invention, but not all embodiments.

[0038] The invention provides a short peptide for treating cardiac fibrosis: GF8-001, the amino acid sequence of which is: Gly-Ala-Lys-Gln-Asn-Glu-Ala-Phe. Through the analysis of the amino acid sequence and its structural domains and important sites of the TRPC6 potential channel, combined with IEDB and PIR software for assignment, evaluation, scoring and FFAS software scoring, the resolution of the integrated PDB crystal is simulated through the template, using AutoDock Vina The software converts the simulated protein crystal structure PDB of TR...

Abstract

The invention discloses a cell membrane transient receptor potential C6 channel immunogenic oligopeptide, a vaccine of the cell membrane transient receptor potential C6 channel immunogenic oligopeptide and a ventricular fibrosis resisting pharmaceutical application of the cell membrane transient receptor potential C6 channel immunogenic oligopeptide, and belongs to the technical field of biological medicine. The short peptide GF8-001 designed, constructed and synthesized by the present invention has an amino acid sequence as shown in SEQ ID NO:1. The short peptide GF8-001 and a carrier protein KLH are coupled through a glutaraldehyde coupling method to form a peptide fragment hapten-coupling protein (namely holoantigen: therapeutic short peptide GF8-001), and the peptide fragment hapten-coupling protein is supplemented with a Freund's adjuvant to induce an organism to generate a target antibody which is lasting and has a very high titer level and aims at TRP6. The target antibody can effectively repress TRPC6 channels on ventricular fibroblasts and inhibit cytoplasm calcium influx of the ventricular fibroblasts through in-vivo and in-vitro level tests, so that proliferation of the ventricular fibroblasts is remarkably inhibited, ventricular muscle fibrillation is prevented, and ventricular remodeling is improved.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and relates to a cell membrane transient receptor potential C6 channel immunogenic short peptide and its vaccine and pharmaceutical application against ventricular fibrosis. Background technique [0002] Ventricular fibrosis is a common pathological change process in the occurrence and development of various heart diseases such as myocardial infarction, cardiomyopathy, and arrhythmia. It is one of the important pathological manifestations of left ventricular remodeling, which can aggravate heart failure and induce malignant arrhythmias. Threat to the life of the patient (Díez Javier, J Am Coll Cardiol, 2020, 75:2204-2218). During the occurrence and development of ventricular fibrosis, fibroblasts proliferate excessively, migrate to the injured area, and transform into cardiac myofibroblasts, and their functions undergo significant changes. The transient receptor potential (TRP) on the cell me...
