Embodiment 1

[0030] 1. Materials and Instruments

[0031] 1.1 Experimental cells

[0032] Grade IV GBM (glioblastoma) human T98G and U118 MG cell lines were purchased from the American Type Culture Center (ATCC, Rockville, USA). Human HEB glial cell line (normal brain glial cell line) was donated by the First Affiliated Hospital of Zhengzhou University.

[0033] 1.2 Experimental Instruments

[0034] Flow cytometer: Beckman Coulter, USA, model: CytoFLEX; enzyme-linked immunofluorescence spot analyzer: CTL, USA, model: CTL-ImmunoSpot Versa; electronic balance (PL602-S, Shanghai Mettler-Toledo International Trade Co., Ltd.); SDS-PAGE electrophoresis instrument: American BIO-RAD, model: Mini-PROTEAN Tetra WBSystem; microplate reader: American Thermo Company, model : MBY001; biological microscope: Japan OLYMPUS Company, model: BX53. Fluorescence microscope: Nikon Corporation, Japan, model: Eclipse E100-LED; confocal microscope: Olympus Corporation, Japan, model: FV1000.

Embodiment 2

[0090] In addition, before this study, we also studied the effect of TBMS2 on the apoptosis of T98G cells in vitro and the combined treatment with TMZ on the expression of MGMT, and found that the reduction effect was not obvious. In addition, no other tupeimoside compounds that can promote and weaken the resistance of glioblastoma to temozolomide and have less adverse reactions to non-tumor cells have been found so far.

[0091] The specific experiments are as follows: the T98G and U118 MG cells pretreated with Control, TMZ, TBMS2, and TBMS2 combined with TMZ (500 μM) in the control group were detected by Western blot experiments. The results showed that MGMT was expressed in each treatment group, and the expression of MGMT There was no significant difference in TMZ, TBMS2, and TBMS2 combined with TMZ, which confirmed that the effect of reducing the expression of MGMT in TBMS2 combined with TMZ was not obvious. For the results, please refer to Image 6 , indicating that TBMS2...

Abstract

The invention discloses application of tubeimoside combined with temozolomide in preparation of a medicine for treating or inhibiting glioblastoma. Under the condition that the temozolomide is adopted to treat glioblastoma, the glioblastoma with temozolomide resistance can be generated, the sensitivity of the glioblastoma with temozolomide resistance to temozolomide can be enhanced by combining tubeimoside with temozolomide for treatment, so that the drug resistance of the glioblastoma to temozolomide is weakened; and the compound has small adverse reaction on non-tumor cells, and has great value in preparation of drugs for treating or inhibiting glioblastoma.

Description

technical field [0001] The invention relates to the field of medicine, in particular to a medicine for treating glioblastoma and its application. Background technique [0002] Glioblastoma (GBM, World Health Organization [WHO] grade IV), accounting for approximately 49.1% of adult malignant primary brain and central nervous system (CNS) tumors, is the most aggressive and refractory high-grade glioblastoma It is one of the most lethal human malignancies with a median survival of only 14.6 months after comprehensive treatment. Currently, glioblastoma is mainly treated in hospitals with tumor resection, adjuvant radiotherapy, and regular temozolomide (TMZ) chemotherapy. However, at least half of glioblastoma patients develop TMZ resistance when treated with temozolomide, leading to compromised therapeutic efficacy. Therefore, it is urgent to develop low-toxicity and high-efficiency therapeutic strategies for TMZ-resistant glioblastoma. [0003] Soil Fritillaria is the dry tu...
