Embodiment 1

[0027] 1. Colchicine induction treatment: take a certain amount of Primula miniature seeds and divide them into 30 groups (treatments), place them in 1.5ml centrifuge tubes respectively, soak in clear water for 24 hours, and then use 0.1g / L, 0.2g / L, 0.5g / L, 1.0g / L, 1.5g / L and 2.0g / L colchicine 1 solutions with six concentration gradients were soaked for 12h, 24h, 36h, 48h and 72h respectively, and a two-factor randomized block design was adopted. Colchicine is easy to decompose when exposed to light, and all treatments are placed in an artificial climate box under dark conditions, and the temperature is controlled at 20±2°C.

[0028] 2. Germination of the treated seeds: After the treatment of Primula chinensis, rinse with water for 3 to 4 times, sow in plug trays in the greenhouse, and use the seeds soaked in water for 48 hours as germination control and greenhouse sowing control. With the increase of colchicine concentration and the prolongation of treatment time, the germin...

Abstract

The invention relates to a process of producing autotetraploid by inducing Primula forbesii with colchicine. It comprises following steps: immersing the seeds of Primula forbesii for 24 hours; then immersing the seeds with colchicine of 0.5-1.5g / l for 36-48 hours in the dark, controlling temperature within 20+-2deg.C; identifying ploidy with morphology and chromosome of cytology. The method is characterized in that tetraploid Primula forbesii is induced with colchicine, the success rate is high, and the effect is good; the genetic stability of tetraploid Primula forbesii F1 generation can be maintained; when compared to diploid Primula forbesii, the tetraploid shows good characters of bigger flowers, much more odorous and thick scape; and it also improves the ornamental value.

Description

technical field [0001] The invention belongs to the field of mutation breeding, and in particular the invention relates to a method for mutagenic tetraploid of Primula miniature and an early identification technology of ploidy. Background technique [0002] Primula forbesii (Primula forbesii) is native to Yunnan and Sichuan in my country. It is a biennial plant of the genus Primulaceae. It is born in ditches, fields, and roadside grass slopes at an altitude of 1500-2800 m. The inflorescences are usually pollinated; umbels 1-2 rounds or more than 2 rounds, 5-15 flowers per round, 5 petals, corolla light red or lavender, lobes apex deeply concave, diameter about 1cm, natural flowering period from December to next March (Chen Fenghuaihe Hu Qiming, 1990), chromosome 2n=18. In 1891, Vilmorin introduced Primula forbesii to France, but now it is rarely seen in European gardens (Richards, 1993). Primula forbesii is in the season of few flowers in winter and spring when it blooms. It ...
