Embodiment

[0019] EXAMPLE Tobacco NaN was carried out according to the following steps 3 Mutation germplasm innovation and variety breeding:

[0020] The first step is the selection of mutagenic materials: select the original tobacco with good appearance quality and internal chemical quality, and resistance to various diseases, but when planted in Guizhou Province, it has poor tobacco leaf identity, large pitch, and high plant height. Air-dried seeds of 11 varieties were used as mutagenic materials. The aim is to select and breed mutants with shortened pitch, moderately reduced plant height and improved tobacco leaf identity through mutagenesis treatment.

[0021] The second step is to prepare NaN3 solution: first mix 4.11ml 0.2M disodium hydrogen phosphate and 15.89 ml0.1M citric acid, among which 0.1M citric acid prepares 1000ml aqueous solution with 21.01g citric acid of one crystal water; 0.2M dihydrogen phosphate Prepare 1000ml aqueous solution with 35.01g disodium hydrogen phosph...

Abstract

The present invention discloses a technique for making tobacco NaN3 mutation germplasm innovation and selecting-breeding variety. Said technique includes the following steps: (1), mutagenic material selection, selecting air-dried seed of tobacco cultivated variety with good comprehensiveness and strong adaptability; (2), preparing NaN3 solution; (3), seed treatment, using NaN3 solution whose concentration is 4mmol / L to treat seed for 6h at 25deg.C; (4), planting and management of M1; (5), planting and selection of M2; and (6), M3 and next-treatment. Said method can greatly raise mutation frequency, and its beeding effect is high.

Description

technical field [0001] The invention relates to a seed treatment method, in particular to tobacco germplasm innovation and variety breeding technology. technical background [0002] Since 1948, Gustafsson et al. used mustard gas to treat barley to obtain mutants; in 1967, Nilan treated barley seeds with diethyl sulfate to breed dwarf, high-yielding varieties. Since then, the research and application of chemical mutagens has gradually developed. Compared with physical mutagens, chemical mutagens have the following characteristics: ① Higher rate of induced mutation and less chromosomal aberration; ② Light damage to processed materials, and some chemical mutagens are limited to certain parts of DNA ③ Most effective chemical mutagens are more harmful to organisms than physical mutagens, and are likely to cause a decline in viability and fertility. In terms of cultivating new plant varieties through chemical mutagenesis, some technical solutions have applied for patents. For exa...
