Embodiment 1

[0047] Example 1 The influence of fermentation time on the production of inulin glycosyltransferase (IFTase)

[0048] According to the fermentation conditions described in the manual, through the detection of the IFTase produced by Arthrobacter aurescens SK 8.001 at different fermentation times, it was found that the IFTase activity was the highest at 72 hours of fermentation, and as the fermentation time prolongs, the IFTase activity decreased slightly, so the fermentation time was determined to be 72 hours about. The effect of fermentation time on inulin-producing glycosyltransferase is shown in Table 1. Enzyme activity is defined as the amount of enzyme required to produce 1.0 μmol of double fructosan III (DFAIII) in 1 minute at a temperature of 55° C. and pH 5.5, which is 1 unit of enzyme activity.

Embodiment 2

[0051] Example 2 The Influence of Inulin Concentration on the Production of Difructose III by Enzymatic Transformation

[0052] According to the enzymatic conversion conditions described in the manual, inulin was prepared into aqueous solutions of different concentrations and reacted for 24 hours at 40°C and pH 5.5. The conversion rate can still be guaranteed to be above 75% under the concentration of the inulin aqueous solution. Table 2 shows the effect of inulin concentration on the enzymatic transformation to produce difructose III.

Embodiment 3

[0055] Embodiment 3 The preparation of double fructose III syrup

[0056] (1) Decolorization

[0057] Add activated charcoal with a solid content of 0.2%-2.0% in the enzyme reaction solution containing double fructose anhydride III, incubate at 80°C for 30 minutes, then filter with diatomaceous earth, and take the decolorization solution;

[0058] (2) concentrated

[0059] Evaporate and concentrate the decolorized solution in a vacuum until the solid content is 60%-95%, and then the double fructose III syrup is obtained.

Abstract

The invention relates to a strain for producing an inulin fructosyltransferase and a method for using the fructosyltransferase to produce a difructose anhydride III, the strain and the method belong to the biotechnology field of foods. The invention relates to an Arthrobacter aurescens SK 8.001 which is sieved from soil, the collection serial No. is CCTCC NO: M 208120, the Arthrobacter is taken as a starting strain, insulin is taken as a carbon source, thereby composing a fermentation culture medium with a nitrogen source and inorganic salt; and the inulin fructosyltransferase is produced by fermentation. The fermentation culture medium takes the insulin and sodium nitrate as the main carbon source and the nitrogen source, and the enzyme activity of the inulin fructosyltransferase in fermentation broth can achieve 2 to 100U/ml through the detection after the fermentation. A catalyst of the inulin fructosyltransferase is added in the insulin solution with 1 percent to 50 percent for producing the difructose anhydride III, the conversion is carried out for 3 to 24h, and the conversation rate can be more than 75 percent. The obtained difructose anhydride III product is safe and reliable, thereby being a functional sweetener with great market potentiality.

Description

technical field [0001] The invention relates to a novel microorganism capable of producing inulin glycosyltransferase and a method for producing inulin glycosyltransferase by cultivating and fermenting it, and using the enzyme to convert inulin to prepare double fructose anhydride III, which belongs to the field of food biotechnology. More specifically, the present invention relates to Arthrobacter aureuscens SK 8.001 derived from soil, which can produce inulin fructosyltransferase, and utilize the inulin fructosyltransferase to convert inulin to generate difructoan III of the following structure ( α-D-fructofuranose-2',1:2,3'-β-D-fructofuranose dianhydride). [0002] Background technique [0003] In recent years, functional food has become the focus of consumers' attention, and its development is the focus of research and development by the majority of food practitioners. The increase and younger age of people with diabetes, obesity and cardiovascular diseases make func...
