Abstract

The invention discloses an in-vitro pollen germination liquid medium of balloon flower (platycodon grandiflorum (Jacq.) A. DC) and a method using same to determine the pollen viability thereof, particularly a method for reliably and effectively determining the pollen viability of the balloon flower through the in-vitro pollen culture of the balloon flower after the in-vitro pollen germination culture system is determined. The in-vitro pollen germination liquid medium is characterized by using distilled water as the solvent with the pH value being 5.8, wherein, the distilled water comprises the following components: ME3, BK, 100g / L sucrose and 150g / L PEG (polyethylene glycol). The method comprises the following steps: evenly scattering the pollen over the in-vitro pollen germination liquidmedium, and culturing at a constant temperature (30 DEG C) under the lighting condition; in-vitro culturing for 1.5h and determining the pollen germination rate and the pollen tube lengths which reflect the pollen viability, particularly, counting the pollen germination rate by the pollen germination standard that the pollen tube length is larger than the pollen particle diameter, and measuring and averaging the pollen tube lengths, wherein, the pollen germination rate reflects the proportion of the viable pollen, and the growth condition of the pollen tube of the in-vitro cultured pollen reflects the physiological condition of the pollen particles.
