Abstract

The invention discloses a method for detecting activity of fluorescein diacetate in bottom sediment. The method comprises the following steps: 1) weighting a bottom sediment sample, adding phosphoric acid buffer solution to shake evenly; adding FDA solution to start a reaction, performing shaking culture for coloration; 2) after the culture, immediately adding chloroform / methanol mixed termination agent to stop the reaction, fully shaking evenly; 3) centrifuging the mixed solution, taking supernatant to detect the absorbance at 490nm; and 4) calculating the amount of fluorescein and performing calculation and unit standardization to the activity of FDA. Proper culture time is selected in the linear region to effectively reduce the detection period; the fluorescein colour reaction can be fast and effectively stopped so as to ensure accurate detection result, fast operation and good reproducibility; the centrifuging time is optimized to ensure to obtain the maximum of the activity of FDA and objectively and scientifically represent the activity of FDA in bottom sediment; the representing unit of FDA is fixed and specified so as to greatly increase the standardization and comparability of the activity data of FDA in bottom sediment.
