Problems solved by technology

However, due to the lack of a comprehensive analysis of the rust resistance genes in the produced varieties in my country, the understanding of the disease resistance of the used varieties is not comprehensive enough, which often leads to the blind promotion and use of wheat varieties, coupled with the emergence of new toxic genes of wheat leaf rust. Disease-resistant varieties continue to lose disease resistance, and the occurrence of wheat leaf rust is becoming more and more serious

Embodiment 1

[0018] Embodiment 1, the acquisition of wheat leaf rust resistance gene Lr24 AFLP-STS marker

[0019] The F of TcLr24 and Thatcher crossed by using the wheat leaf rust resistance gene Lr24 low toxicity, high toxicity to the susceptible parent Thatcher monospora line (BGQQ and SHRT) of wheat leaf rust resistance gene Lr24 1 and F 2 F 1 Group 10 strains, F 2 Population 141 strains, the result is F 1 All disease resistant, F 2 The resistance and sensitivity conformed to the segregation ratio of 3:1, as shown in Table 1, indicating that the resistance of TcLr24 to BGQQ and SHRT was controlled by a dominant single gene.

Abstract

The invention discloses a method and a special primer for screening a wheat leaf rust resistance gene Lr24. The special primer is a primer pair consisting of nucleotide sequences in a sequence 1 and a sequence 2 in a sequence list. The method for screening wheat containing the leaf rust resistance gene Lr24 is to perform polymerase chain reaction (PCR) amplification by using the genome DNA of a wheat variety to be tested as a template under the guidance of the primer pair consisting of the nucleotide sequences in the sequence 1 and the sequence 2 in the sequence list according to claim 1, wherein the product of the amplification contains wheat which has a strip size of 212bp and has the Lr24 gene. A method for detecting the amplification products comprises the steps of performing 2.0 percent agarose gel electrophoresis on the amplification products and developing by using an electronic beam (EB) or Goldview. The method and the special primer for screening the wheat leaf rust resistance gene Lr24 are expected to play an important role in the wheat leaf rust resistance breeding.

Description

technical field [0001] The invention relates to a method for screening wheat leaf rust resistance gene Lr24 and special primers thereof. Background technique [0002] Wheat leaf rust (Puccinia triticina f.sp.tritici) is a worldwide disease that can cause yield losses of 40% or more under favorable conditions (from Knott D R. The wheat rust breeding forresistance. Monographs on Theoretical and Applied Genetics , 1989). In my country, wheat leaf rust used to occur mainly in Southwest China and parts of the Yangtze River Basin, such as Guizhou, Jiangxi and other provinces. In recent years, the occurrence of wheat leaf rust has become increasingly serious in North China, Northwest China and Northeast China. The use of leaf rust-resistant varieties is an economical, effective and quick way to reduce the damage of leaf rust. Strengthening the research on the disease resistance of wheat varieties and their genetic resources is very important for the continuous control of diseases...
