Problems solved by technology

The existing macroporous resin column chromatography technology adopts normal pressure technology, the resin particles used are coarse and uneven, and the column efficiency is low. The color value of the separated gardenia yellow product is generally below 400, and it is difficult to reach more than 500, and the OD value is generally high. 0.4, unable to meet the quality requirements of exporting to Japan with an OD value of less than 0.2

Embodiment 1

[0016] 1. Extraction: Crush 1 kg of dried gardenia fruit to 10-40 mesh, soak in 8-15 kg of pure water for 1 hour at room temperature, then infiltrate at an appropriate flow rate, share 25-35 kg of pure water for percolation and extraction, 4 -8 hours percolation extraction is complete, to obtain the extract A.

[0017] 2. Microfiltration: pass the extract A through a 0.2-1 micron ceramic membrane to obtain a clarified filtrate B.

[0018] 3. Sample loading and adsorption: Use 3 sections of ZT-02 macroporous adsorption resin columns, each column volume is 70 ml. Put the clarified filtrate B on the first column for adsorption until the effluent is reddish, connect the second column at the end of the first column, and continue to absorb until the effluent of the second column is reddish. String the column C at the end of the second column until the effluent from column C is reddish, then remove the column C. At the end of the second column, connect column D again, and handle th...

Abstract

The invention relates to a process for extracting gardenia yellow from gardenia. The process comprises the following steps of: crushing raw materials, performing water diacolation for extracting, performing micro-filtration, performing macroporous resin column chromatography, separating and purifying, concentrating and drying products and activating and regenerating columns, wherein in the step of performing water diacolation for extracting, dry gardenia fruits are crushed into large granules, and the large granules are soaked in pure water and are subjected to the diacolation at normal temperature to be extracted; in the step of performing micro-filtration, a water extracting solution is subjected to the micro-filtration by using 0.2 to 1 micrometer of ceramic membrane to obtain a completely-clarified filtrate; in the method for performing loading adsorption continuously, three sections of columns are connected in series to perform the loading adsorption, and the third section of column is separated and purified further; and in the step of activating and regenerating columns, the efficient macroporous resin columns are separated continuously under high pressure and are activated and regenerated on line. According to the process, a constant pressure technology is upgraded to a pressurization technology, so that the adsorptive capacity and separation efficiency of the resin columns are improved; and according to the high-end gardenia yellow products, the color scale of the prepared gardenia yellow is over 500, an optical density (OD) value is below 0.2, and the content of geniposide is less than 1 percent by high performance liquid chromatography (HPLC) detection on the basis of the color scale of 500.

Description

technical field [0001] The invention relates to a process for extracting gardenia yellow pigment from gardenia jasminoides, which adopts percolation extraction, microporous filtration, high-efficiency macroporous resin column chromatography and other related technologies, and belongs to the field of extraction, separation and purification of natural products. Background technique [0002] The water-soluble natural yellow pigment extracted from the mature fruit of Gardenia is called Gardenia Yellow, and its main components are crocin and crocetin, which are widely used as food additives in the coloring of food, medicine and cosmetics at home and abroad. Gardenia yellow is usually extracted with water and ethanol, and can be extracted by different processes such as thermal extraction, ultrasonic extraction, microwave extraction, continuous extraction, countercurrent extraction, and osmotic pressing. The gardenia fruit needs to be crushed before extraction, because the gardenia...
