Embodiment Construction

[0024] Take 5.0 kg of raw Spirulina medicinal materials, add 20 L of 95% industrial ethanol, and soak at room temperature for seven days to remove fat-soluble small molecules. Pour off the ethanol, put the solid matter in a ventilated place to dry, and then extract with boiling water, adding about 30 L of water each time, and the extraction time is 4 hours. Use the phenol-sulfuric acid method to detect the sugar content of the extract until the sugar reaction is not obvious. , a total of 5 extractions. Each extract was filtered and combined, heated and concentrated to a small volume, dialyzed against flowing water for 2 days, the dialyzed liquid was concentrated and centrifuged, 1:1 (v / v) was added with 30% trichloroacetic acid solution to remove protein, and the React at 4°C for 4 hours, add 10% NAOH solution to neutralize until the pH value is about 7, then tie a bag and dialyze against flowing water for 48 hours, concentrate the inner liquid of the dialysate and centrif...

Abstract

The invention relates to spirulina phatensis polysaccharide and an extraction method thereof. The extraction method comprises the following steps of: immersing spirulina which is taken as a raw material at room temperature for seven days by using industrial ethanol, pouring ethanol, airing solid, carrying out extraction for many times by using boiling water, dialyzing fluid water for two days, after concentrating intra-dialysate, centrifuging, adding a trichloroacetic acid solution to eliminate protein, neutralizing by adding an NAOH solution till the pH value is 6.5-7.5, after dialyzing and concentrating fluid water, centrifuging, adding triploid product of industrial ethanol into supernate, standing for one night, centrifugally collecting precipitation, and drying after washing through absolute ethyl alcohol and acetone respectively so as to obtain crude aqueous extraction spirulina phatensis polysaccharide; and carrying out solid residue alkali extraction to obtain crude alkali extraction spirulina phatensis polysaccharide, and separating and eluting crude polysaccharide so as to obtain pure polysaccharide. The chemical structure of the spirulina phatensis polysaccharide is characterized in that a main chain is composed of 1,4-connection alpha-D-glucose; average every eight main chain residues have one branch; substitution is at the sixth position of a branch point; and the branch is composed of one or two 1.6 connection glucose. The extraction method of the spirulina phatensis polysaccharide disclosed by the invention is simple and convenient and has good reproducibility.

Description

technical field [0001] The invention relates to a spirulina polysaccharide and an extraction method thereof. Background technique [0002] Spirulina is a class of lower plants, belonging to Cyanophyta, Oscillatoriaceae. Like bacteria, they do not have a real nucleus in their cells, so they are also called cyanobacteria. The cell structure of cyanobacteria is primitive and very simple. It grows in the water body, and its morphology can be seen as a spiral filament under the microscope. There have been reports on the antitumor activity of spirulina polysaccharides, but most of them are crude polysaccharides. There is no report on which polysaccharide the antitumor activity is derived from and what its clear structure is. Contents of the invention [0003] The purpose of the present invention is to provide a kind of spirulina polysaccharide and its extraction method. [0004] Technical scheme of the present invention is: [0005] The extraction method of spirulina polysa...
