Problems solved by technology

[0006] 1. The method of taking tissue from a living body must be adopted. If the animal is dead, extensive and diffuse thrombus will form in the blood vessels in the liver after about 10 minutes, making it difficult for the detergent to reach the decellularized site. The detergent only acts on the lipids on the surface of the cell membrane. quality components, so the decellularization efficiency is not high, and the decellularization effect cannot be achieved
[0007] 2. The method of taking tissue from a living body has high requirements on the source of liver donors. If it is used in clinical practice, the source of liver stent donors will be limited, which is difficult to promote and has little clinical practicability

Embodiment 1

[0015] A liver tissue decellularized liquid is a new type of decellularized liquid made by introducing two components into the animal whole liver decellularized technology. The decellularized liquid is composed of 0.01% recombinant human tissue plasminogen activator (rt-PA ) and 2% tween80, prepared with sterile saline.

[0016] Specific operation:

[0017] 1. Whole liver acquisition:

[0018] After the rat was euthanized, the abdomen was cut longitudinally to expose the rat liver. The proximal vena cava, portal vein and hepatic artery were marked with a 20G cannula (BD Intima) and cut off. After the falciform ligament and heart-shaped ligament were separated and transected, the liver was gently moved to expose the inferior vena cava to the field of view. Ligate and separate the distal vena cava and any appendages adhering to the liver.

[0019] 2. Decellularization preparation:

Embodiment 2

[0033] A liver tissue decellularized liquid is a new type of decellularized liquid made by introducing two components into the whole animal liver decellularized technology. The decellularized liquid is composed of 0.02% recombinant human tissue plasminogen activator (rt-PA ) and 1% tween80, prepared with sterile saline.

[0034] Specific operation:

[0035] 1. Whole liver acquisition: the same as in Example 1.

[0036] 2. Decellularization preparation:

[0037] (1) Decellularized solution (proportion: 0.02% rt-PA, 1% tween 80) was injected through the portal vein at 37°C, and the hepatic artery cannula and proximal vena cava were clamped for 30 minutes. Then clamp the hepatic artery cannula, open the proximal vena cava, connect the portal vein to the peristaltic pump, and perfuse the portal vein with decellularized solution at a rate of 4ml / min.

[0049]A liver tissue decellularized liquid is a new type of decellularized liquid made by introducing two components into the animal whole liver decellularized technology. The decellularized liquid is composed of 0.05% recombinant human tissue plasminogen activator (rt-PA ) and 0.5% tween80, prepared with sterile saline.

[0050] Specific operation:

[0051] 1. Whole liver acquisition: the same as in Example 1.

Abstract

The invention discloses a liver tissue cell removal solution. The novel cell removal solution prepared from two components is introduced into a whole animal cell removal technology. The cell removal solution is prepared from 0.01 to 0.05 percent of recombinant tissue plasminogen activator (rt-PA) and 0.5 to 2 percent of tween-80 by using sterile normal saline. By using the strong thrombolytic effect of the rt-PA and the solubilization and red blood cell destruction effects of the tween-80, the liver tissue cell removal solution is relatively easy to prepare and implement, high in cell removal efficiency and low in requirement for warm ischemia time of the liver, and greatly improves the potential probability of a cell removal liver scaffold in clinical application.

Description

technical field [0001] The invention relates to a biological scaffold, in particular to a liver tissue decellularized liquid which can be used as a biological scaffold material in liver tissue engineering. Background technique [0002] End-stage liver diseases such as severe liver failure and cirrhosis have seriously affected people's quality of life and become one of the diseases with the highest mortality in the world. Currently, liver transplantation is recognized as the only effective treatment. However, the shortage of donor livers has limited the promotion of this technology, and many patients have lost the best opportunity for treatment. [0003] Tissue engineering is a subject that comprehensively uses the basic principles, basic theories, basic technologies and methods of biology and engineering to study how to construct biologically active tissues and organs under in vitro conditions to replace damaged tissues and organs. An ideal tissue engineered tissue or orga...
