Problems solved by technology

This type of method is time-consuming, labor-intensive, easily affected by the operation method, and has low sensitivity. Under the current technical conditions, the microorganisms that can be isolated by culture only account for about 0.1%-1% of the total number of bacteria, and can only be qualitatively detected. Bacteria that can be cultured , cannot identify unknown bacteria, so it cannot correctly reflect the quantity and diversity of intestinal flora

Embodiment 1

[0055] The NF-κB-luc transgenic mice were fed in groups, 3 mice in each group, and the group feeding method was as follows:

[0056] The first group (Ctrl group): fed with normal feed (maintenance feed for rats and mice from Beijing Huafukang Biotechnology Co., Ltd., product code 1022);

[0057] The second group (GM group): feed mixed mouse diet (mix 1 volume part of transgenic corn powder with 3 volume parts normal feed to obtain mixed mouse diet).

[0058] 2. After 10 weeks of group feeding, the feces of the mice were taken, and the total DNA was extracted using the feces sample DNA extraction kit (Kangwei Biotechnology Company).

[0059] 3. Take the total DNA obtained in step 2 and crush it with a DNA ultrasonic pulverizer (Covaris S220) to obtain DNA fragments of 300-800 bp.

[0060] 4. Use the Roche454GS Junior sequencer (Roche) and the matching GS FLX reaction system kit to perform high-throughput sequencing of bacterial 16S rRNA on the DNA fragments obtained in step 3 ...

Abstract

The invention discloses an auxiliary method for detecting transgenic plant components in foods. The method comprises the following steps: 1, separating NF-kB-luc transgenic mice into two groups, wherein the first group is fed by common fodder while the second group is fed by to-be-detected fodder which is a mixture of to-be-detected foods and common fodder; 2, after 10 weeks, taking manure of the mice and extracting total DNA; 3, taking and breaking the total DNA to obtain a DNA segment of 300-800 bp; 4, taking the DNA segment for high-throughput sequencing of 16 S rRNA; 5, based on the results, obtaining composition of intestinal flora, wherein if the relative abundance of firmicutes in the manure of the second group of mice is obviously higher than that of the first group, and statistical difference appears, the to-be-detected foods contain the transgenic plant components. The method is simple and convenient in operation, low in cost and simple in result judgment, and has very high promotion and application values.

Description

technical field [0001] The invention relates to a method for assisting detection of whether genetically modified plant components are contained in food. Background technique [0002] Genetically modified organisms are a concept accepted by all countries in the United Nations Convention on Biosafety Protocol, called "Living Modified Organisms", LMOs for short, or "Genetically Modified Organisms", GMOs . LMOs or GMOs refer to living organisms with novel combinations of genetic materials obtained by means of modern biotechnology. In fact, foreign DNA is introduced into the genome of organisms, causing genetic changes, and organisms that change the genetic composition are called genetically modified organisms. [0003] Since 1996, genetically modified plants have been popularized on a large scale, such as genetically modified soybeans, genetically modified corn, and genetically modified tomatoes. In the world, the United States is the country with the fastest development of g...
