Problems solved by technology

Due to the long incubation period of the pathogenic bacteria of Huanglongbing in citrus and the lack of effective drug control, once the disease occurs, the final result is death, which has caused many local excellent varieties or excellent breeding materials and germplasm resources to be on the verge of extinction.
[0004] After searching, at present, there is no relevant literature report on the rapid conservation of excellent germplasm resources or varieties of citrus that are endangered due to the serious harm of Huanglongbing

Embodiment Construction

[0017] The present invention will be described in detail below in conjunction with specific embodiments.

[0018] (1) Heat treatment of citrus rootstocks: put the susceptible and healthy citrus rootstock seedlings in an artificial climate chamber for heat treatment, 10 hours a day at 45°C, and then maintain 35°C for 14 hours for 5 days, and then treat The rootstock seedlings were kept in an insect-proof greenhouse. After 3 weeks, the treated rootstock seedlings resumed growth. After 4 months and 8 months after heat treatment, the number of pathogenic bacteria of Huanglongbing and the number of heat-treated rootstock seedlings were detected by quantitative PCR. Effect. ① Extraction of DNA of pathogenic bacteria: extraction with Qiagen plant DNA extraction kit (DNeasy Plant Mini Kit, Cat. 69106, Qiagen, MO, USA). Take the leaves of the rootstock seedlings, tear off the midrib, put about 200mg of the midrib into a 2.0ml centrifuge tube, add AP1 buffer and RNAase, break it up wit...

Abstract

The invention relates to a method for cultivating citrus germplasm infected with citrus vein phloem degeneration. The method comprises the steps of cultivation and heat treatment of rootstock, collecting of scions, treating drugs, grafting, managing and detecting. According to the method, heat treatment is conducted on rootstock seedlings, the rootstock without the citrus vein phloem degeneration is cultivated, drug treatment is conducted on citrus branches which are infected with the citrus vein phloem degeneration while still have vitality, and the scions are taken from the citrus branches after drug treatment and grafted on the rootstock without the citrus vein phloem degeneration. Samples are collected in four months, eight months and twelfth months after grafted seedlings are survived, and PCR detection and morbidity situation survey are conducted. The result shows that pathogenic bacteria of the citrus vein phloem degeneration of infected scions of citrus elite germplasm or variety can be completely eliminated, the recovery and cultivation effect of infected germplasm resources can be visually and accurately evaluated, and the citrus germplasm resources can be quickly protected and cultivated.

Description

technical field [0001] The invention relates to a method for conserving fruit tree germplasm infected with diseases, in particular to a method for conserving citrus germplasm infected with Huanglongbing, belonging to the field of agricultural science. Background technique [0002] Citrus Huanglongbing is a devastating, contagious and quarantine disease caused by Ca. Liberibacter. Occurs and is endemic in more than 40 countries and regions including Asia, Africa, and South America, and is mainly transmitted through citrus psyllid transfer and feeding, diseased seedlings and scions. Usually new trees will die within 1 to 2 years after being infected, and old trees will die or lose fruiting ability within 3 to 5 years after being infected. In severe cases, the garden may be destroyed. For example, citrus huanglongbing was first discovered in Florida, USA in 2005. In just 5 years, it has spread to all 34 citrus producing counties, causing economic losses as high as 4.6 billion ...
