Problems solved by technology

In the breeding work of freshwater shrimp, in order to avoid the reduction of offspring viability and germplasm degradation caused by inbreeding, the paternity identification of freshwater shrimp is particularly urgent, but there is no effective method to identify the parent-child relationship of freshwater shrimp. The method of the family line is to bind the eye-stalk mark on the eye stalk, which not only consumes manpower and material resources, but also the binding is not suitable for juvenile shrimp, and it still needs to be reared in separate ponds for a period of time before the juvenile shrimp grows large enough. The method of identifying the parent-child relationship of shrimp has extremely important theoretical and production significance
There is no report on the application of microsatellite markers in the family identification of freshwater shrimp

[0019] (1) Breeding of freshwater shrimp family

[0020] 10 female parents and 10 male parents were selected from the natural population of freshwater shrimp (the green shrimps used in the experiment were all wild adult freshwater shrimps, all collected from Wuxi Lake District of Taihu Lake (120°13'44"E, 31°28' 22"N)), through one male to one female breeding, in order to prevent the male shrimp from attacking the female shrimp and causing the female shrimp to die, each female shrimp is fed in a separate tank, and after the female shrimp molts, a male shrimp The shrimps were put into the water tank to complete the mating. After the female shrimps were observed to lay eggs, the male shrimps were sampled and stored in 95% alcohol at -20°C. After the eggs hatched, the female shrimp was sampled, stored in 95% alcohol, and kept at -20°C. Finally, 3 families were obtained. The offspring of shrimp seedlings were mixed after dyeing and sampled on the 20th day, and a total of 126 indiv...

Abstract

The invention relates to a primer set and method for identifying macrobrachium nipponensis families by using microsatellite markers and belongs to the technical field of macrobrachium nipponensis breeding. The method comprises the following steps: breeding the macrobrachium nipponensis families artificially; collecting macrobrachium nipponensis samples to be identified, and extracting genome DNA; amplifying genome DNA by carrying out polymerase chain reaction (PCR) amplification on selected 12 pairs of microsatellite primers based on the obtained genome DNA as a template; carrying out electrophoretic detection on the PCR amplification product; and processing the electrophoretic result digitally, and carrying out parental test by using genetic analysis software. According to the invention, parental test is carried out on macrobrachium nipponensis so as to distinguish different families and reduce inbreeding existing in breeding of macrobrachium nipponensis groups to facilitate family breeding, thereby breeding process and improving the breeding effect.

Description

technical field [0001] The invention relates to a primer group and a method for using microsatellite markers to identify freshwater shrimp families, and belongs to the technical field of shrimp breeding and molecular markers. Background technique [0002] Green shrimp, scientific name Macrobrachium nipponense (Macrobrachium nipponense), commonly known as river shrimp, has the advantages of high fecundity, strong adaptability, wide appetite, delicious meat, and can be marketed all year round. At present, the annual output of cultured freshwater shrimp in the country exceeds 230,000 tons, and the annual output value is nearly 20 billion yuan. Freshwater shrimp farming has played a pivotal role in my country's aquaculture. However, most of the current cultured freshwater shrimp are the offspring of wild species that have not been systematically genetically selected and bred in ponds for multiple generations. The degradation phenomenon has seriously restricted the development o...
