Abstract

The invention relates to the field of cytobiology, and discloses a method for separating and purifying Schwann cell from peripheral nerves of an adult macaque. The method comprises the following steps of 1. obtaining of nerves and in-vitro initial denaturation: under the non-bacteria condition, obtaining the sural and peroneal nerve sections of the adult macaque, removing epineurium, and adopting a SCCM (sertoli's cell culture medium) initial denaturation culture medium containing composite factors to culture, so as to obtain the sural nerves of the adult macaque; 2. primary culture of cells: mixing NB4 and dispase to digest nerves, centrifuging, resuspending in a culture dish of laminin containing CCM (complete culture medium), and incubating; 3. purification of Schwann cells: after incubating of the cells, adding into dispase, digesting, collecting a cell suspension liquid, and incubating in a purifying culture liquid containing Ara-C; then, purifying, centrifuging, and planting into a growth culture medium containing the SCCM. The method has the advantage that in the initial denaturation process, the cell proliferation factors are added, so that the output of cells is improved, the purification time is shortened, and the Schwann cell of the adult macaque with high purity is obtained.
