Embodiment Construction

[0033] The present invention will be described in detail below with reference to the drawings and specific embodiments, but it should be understood that the protection scope of the present invention is not limited by the specific embodiments.

[0034] The invention provides a gene fragment, the nucleotide sequence of which is shown in SEQ ID NO.1.

[0035] The present invention also provides a vector pPlasmid-Clearance, wherein the vector pPlasmid-Clearance inserts a gene whose nucleotide sequence is shown in SEQ ID NO. The nucleotide sequence of the conjugative transfer vector pEF01 is shown in SEQ ID NO.2, and the resistance gene region refers to the nucleotide sequence between 3730-5249 of the conjugative transfer vector pEF01, Specifically follow the steps below:

[0036] Step 1, artificially synthesizing a gene fragment whose nucleotide sequence is shown in SEQ ID NO.1;

[0037] The gene segment whose nucleotide sequence is shown in SEQ ID NO.1 is divided into figure 1...

Abstract

The invention discloses a gene fragment which has a nucleotide sequence as shown in SEQ ID NO.1; the invention also provides a vector pPlasmid-Clearance, wherein the vector pPlasmid-Clearance is constructed by inserting the gene fragment having the nucleotide sequence as shown in SEQ ID NO.1 into a resistance gene region of a conjugal transfer vector pEF01 through a homologous recombination method; the conjugal transfer vector pEF01 has the nucleotide sequence as shown in SEQ ID NO.2. The gene fragment and the vector pPlasmid-Clearance can reverse the drug resistance of drug resistant bacteria of polymyxin drug-resistant gene mcr-1 in bacterial flora of environments and animals, can block the spread of polymyxin drug-resistant gene mcr-1, and have great application potential in the prevention and clinical treatment of bacterial diseases.

Description

technical field [0001] The invention belongs to the technical field of genetic engineering, and specifically relates to a gene segment, a carrier pPlasmid-Clearance and applications thereof. Background technique [0002] The widespread spread of drug-resistant bacteria has seriously threatened the health of humans and animals. Bacterial drug resistance can be divided into two forms according to its causes: one is natural drug resistance, also known as inherent drug resistance, which is determined by the drug resistance-related genes on the bacterial chromosome and passed vertically between bacterial generations, also known as Natural drug resistance; the second is acquired drug resistance, which is mainly acquired. With the long-term and large-scale use of antibacterial drugs, the plasmid containing the drug resistance gene is transferred horizontally between bacteria through the conjugation reaction , mediate the recipient bacteria receiving the plasmid to produce drug-res...
