Embodiment 1

[0069] Find mulberries with typical mulberry sclerotinia symptoms in the diseased mulberry garden, collect and cut the diseased area in the mulberry fruit, and the cut materials are fully ground with liquid nitrogen, and the total DNA is extracted using Kangwei Century Co., Ltd. The Fungal DNA Extraction Kit, specifically according to its operating instructions, the total DNA after extraction was stored at -20°C. According to the Illumina library construction process, the total DNA was constructed as a paired-end high-throughput sequencing library with a fragment size of 450bp. The constructed DNA library was subjected to high-throughput sequencing using an Illumina Hiseq2500 sequencer. A total of 9.03M pairs of sequencing fragments were measured and sequenced. The read length is 125bp at both ends, and the total sequencing data volume is 2.26Gb.

[0070] Since the mulberry material of mulberry was included in the DNA extraction process, in order to reduce the influence of mul...

Abstract

The invention discloses a high-throughput mulberry pathogenic bacteria identification and species classification method. The method comprises the following steps that diseased mulberries are collected; the total DNA of the diseased mulberries is extracted; an Illumina DNA library is created; Illumina high-throughput sequencing is carried out; a mulberry genome sequence in sequencing data is removed; microbial genome sequences are assembled; complete ribosomal DNA sequences are assembled; microbial ribosomal DNA sequences are screened and labeled; the ribosomal DNA sequences are comparatively analyzed to classify species, and thereby the mulberry pathogenic bacteria identification and species classification are fulfilled. A result shows that three species of fungi are identified in total when the method disclosed by the invention is applied to carry out the identification of pathogenic bacteria of popcorn disease and species classification, wherein the Ciboria pathogenic bacteria has the highest relative abundance, hereby the pathogenic Ciboria shiraiana is determined as Ciboria, and according to a comparison result, the pathogenic Ciboria shiraiana is determined as Ciboria carunculoides. Most of the species are phytopathogenic bacteria, and can lead to symptoms, such as mummification and swelling, appearing on fruits and seeds of plants, which are identical with the symptoms of the popcorn disease.

Description

technical field [0001] The invention relates to the technical field of plant pathogenic bacteria identification and species classification, and more specifically, relates to a method for high-throughput identification and species classification of mulberry pathogens. Background technique [0002] In recent years, with the diversified development of sericulture and the rise of the mulberry industry, mulberry not only uses mulberry leaves as the feed for silkworms, a special economic animal in the sericulture industry, but also because of the pharmacological and nutritional value of the mulberry tree itself, making mulberry multi-purpose Resource utilization becomes more abundant. The whole body of the mulberry tree is full of treasures. Mulberry leaves, mulberries and mulberry branches are the main harvests for planting mulberry at present. ", since ancient times, it has been a kind of diuretic, health care, and refreshing fresh fruit that people often use. It can be made in...
