Problems solved by technology

So far, some foreign and domestic provinces have relatively complete cultivation management theory and technology. The main propagation method is cuttings, which cannot meet the requirements of the market, and asexual reproduction will lead to virus accumulation and quality degradation.
Previous studies on the in vitro culture of Panax notoginseng have been carried out, including Jiang Xianghui and Xiao Shulin using the young leaves and stems with buds of Panax notoginseng as explants, and establishing a tissue culture and rapid propagation system by means of callus. It lays the foundation for the research of Panax notoginseng in this field, but it takes a certain amount of time for the callus to differentiate into seedlings, and it is also prone to mutation. As a commercial promotion, there are certain restrictions

Embodiment 1

[0023] 1. Acquisition of Rattan Notoginseng Explants

[0024] Take Panax notoginseng from about 10cm above the root to about 10cm away from the top of the stem, cut into 2-3cm sections of stems with leaf buds as explants, and rinse them with tap water for 30 minutes. First sterilize with 75% alcohol for 30 seconds; rinse twice with sterile water; then sterilize the stem segment in 0.1% mercuric chloride for 8 minutes; then remove the explants and soak them in sterile water for 2 minutes, and then use Rinse with sterile water 5 times respectively. Finally, sow the stems of Panax notoginseng into MS medium to cultivate adventitious buds, and place them under white light conditions with light intensity of 1500Lx, light time of 14h / d, and culture temperature of 25±2°C for 15 days.

[0025] 2. Primary culture

[0026] Take adventitious buds of the same size from test tube materials with consistent growth as explants, inoculate them in MS medium, and then place them under white li...

Abstract

The invention provides a method for establishing an anredera cordifolia in-vitro regeneration system. The method comprises the following steps: (1) establishing a sterile system, wherein a best effect can be achieved by sterilizing an anredera cordifolia stem section which is 100cm below the top end for 8min by utilizing HgCl2; (2) carrying out the primary culture: MS is adopted as a basic culture medium, so that the stretching growth of the anredera cordifolia stem section under the white light condition can be facilitated; (3) carrying out the multiplication subculture: a multiplication effect of an anredera cordifolia test-tube seedling on a culture medium of MS+6-BA 1.6mg / L+NAA 0.6mg / L is best, and a multiplication coefficient can reach 15.31+ / -0.28; and (4) carrying out the rooting culture: the rooting effect of MS+1.5mg / LIBA culture medium for the induced anredera cordifolia test-tube seedling under the white light condition is relatively good, and the rooting rate of the test-tube seedling reaches 94.44 percent; and (5) transplanting: after the seedling exercise for 4 days, the test-tube seedling is transplanted onto a vegetable garden soil culture medium, and the survival rate can reach up to 95 percent.

Description

technical field [0001] The invention relates to a method for establishing a rattan notoginseng in vitro regeneration system, belongs to the technical field of plant tissue culture, and is mainly used in the propagation of rattan notoginseng seedlings. Background technique [0002] Panax notoginseng ( Boussingaultia gracilis Miers var. Baselloides Bail . ), for the Laceaceae ( Basellaceae ) Boussingaultia (Boussingaultia H.B.K.) plant, formerly known as heart-shaped Luokui potato, also known as Luokui potato, Yang Luokui, Chuanqi, Tuchuanqi, Fujiko Sanqi, Xiaonian medicine, Tu Sanqi, Madeira rattan, Qianjinzhu, Zhongzhilian, Tianzhi, etc., are native to Brazil and are cultivated in Jiangsu, Zhejiang, Fujian, Sichuan, Guizhou, Yunnan and other places in China. Rattan notoginseng is a perennial twining herbaceous vine, with fleshy stems, round to oval, light purple-red in the initial stage, strong twining, and adventitious roots between nodes. [0003] Because rattan no...
