Abstract

The tissue culture method comprises the following steps: firstly, selecting a female parent material; secondly, treating an explant: taking veins of female parent tender leaves as the tissue culture explant; washing the leaves with tap water after cutting from the female parent material; successively soaking with ethanol, flushing with sterile water, soaking with hydrogen peroxide and flushing with sterile water; cutting the veins along the edges of the veins; thirdly, starting culture: horizontally inoculating the cut veins into a starting medium and putting into a culture chamber for culturing; fourthly, carrying out differentiation culture on callus:after starting culture is ended, forming the callus at the edges of the veins and transferring the callus into a callus differentiation medium; fifthly, carrying out induced culture on the adventitious buds: after differentiation culture of the callus is ended, the callus is enlarged; transferring the callus into the medium for induced culture of the adventitious buds and inducing and differentiating the adventitious buds on the callus.
