Embodiment 1

[0117] Embodiment 1, the method for identifying wheat grain feruloyl arabinoxylan (Feruloyl arabinoxylan, FAX) content

[0118] 1. Molecular markers for identification of feruloyl arabinoxylan content in wheat grains

[0119] 1. There are two allelic variant forms of the TaBahd-A1 gene in wheat: one such as the 19th-1408th position from the 5' end in SEQ ID No.5 (the 445th-568th position of SEQ ID No.5 is inclusive Subsequence) nucleotide molecule shown in (named allele TaBahd-A1a); Another kind is as shown in SEQ ID No.6 from the 19th-1427th position of 5' end (the first 445-587th of SEQ ID No.6 position is the intron sequence) nucleotide molecule (named TaBahd-A1b).

[0120] 2. Design specific primer pair 1 and primer pair 2 based on the two allelic variant forms of the TaBahd-A1 gene as follows:

[0121] The upstream primer of primer pair 1 (SEQ ID No.1): 5'-TGCTCCCGCTGTCCTCC-3';

[0122] The downstream primer of primer pair 1 (SEQ ID No.2): 5'-CGGGTGTTGTTTCTCTTTTG-3'; ...

Embodiment 2

[0142] Example 2, verification of molecular markers

[0143] Experimental materials: 141 Chinese winter wheat main varieties (as shown in Table 1), the public can obtain from the National Germplasm Bank of the Institute of Crop Science, Chinese Academy of Agricultural Sciences.

[0144] The method for identifying the content of feruloyl arabinoxylan in wheat grains described in Example 1 was used to verify 141 main Chinese winter wheat varieties. At the same time, various wheat varieties were planted in Anyang, Henan, and the FAX content of the wheat grains was tested after harvesting. The FAX content of each wheat material was tested twice. If the error of the two test results exceeds 10%, a third repeated test is required. The results are averaged.

[0145] The method for determining the FAX content of the grain is as follows:

[0146] 1. Remove protein and starch from wheat flour

[0147] (1) Weigh 0.2 g of fresh whole wheat flour into a 2.0 mL centrifuge tube, and add 1...

Abstract

The invention discloses a molecular marker relevant to the wheat grain ferulic acid arabiaxylan (FAX) content. The invention provides a set primer for identifying the content of the wheat grain ferulic acid arabiaxylan content. The molecular marker consists of a primer pair 1 and a primer pair 2, wherein the primer pair 1 consists of single-chain DNA shown as SEQ ID No. 1 and SEQ ID No. 2; the primer pair 2 consists of single-chain DNA shown as SEQ ID No. 3 and SEQ ID No. 4. The molecular marker can be used for fast identifying TaBahd-A1a and TaBahd-A1b allelic variation and the relevant relationship between the allelic variation and the wheat FAX content; the wheat variety with high FAX content is screened out; the culture step of the new variety of the high-quality wheat is accelerated;in addition, the important theoretical significance and economic values are realized on the auxiliary selection of high-FAX-content wheat variety by using the molecular marker.

Description

technical field [0001] The invention belongs to the field of biotechnology, and in particular relates to a molecular marker for identifying the content of feruloyl arabinoxylan in wheat grains and an application thereof. Background technique [0002] The content of feruloyl arabinoxylan has an important influence on the processing and nutritional quality of flour products. Although the content of Arabinoxylan (AX) in flour is small (2.3-6.8%), it is an important component of dietary fiber in wheat and has an important impact on the processing and nutritional quality of flour and its products. In addition to protein and starch, it has become the most concerned wheat component, and thus has become a research hotspot related to health. The basic structure of AX is composed of xylan main chain and α-L-arabinofuranosyl (Ara) side chain, and the xylan main chain is composed of β-D-xylopyranose residue (Xyl) Linked by β-(1→4) glycosidic bonds; Feruloyl acid (FA) and AX pass throu...
