The method comprises the following steps: adding an ammonia nitrogen solution or a nitrite nitrogen solution into a pretreated dialysis bag, adding the ammonia nitrogen solution or the nitrite nitrogen solution of the same concentration and an EPS solution have the same concentration and volume in a pure substrate solution into another dialysis bag, clamping the openings of the dialysis bags to prevent water leakage, adding the same weights to make the dialysis bags suspend or completely immerse in an external anaerobic ammonia-free water environment, placing at 20 to 35 DEG C for dialysis, measuring the concentration of the ammonia-nitrogen solution or the nitrite nitrogen solution in the external solutions of the dialysis bags every 1 hour, and fitting dialysis curves of two dialysis experiments with Origin8.0, wherein the adsorption amount of the ammonia nitrogen solution or the nitrite nitrogen solution by EPS is the difference between final external dialysis concentration fitted by a pure substrate and final external dialysis concentration fitted after adding of the EPS solution.
