Problems solved by technology

From the acquisition of biological samples to RNA extraction, library construction and sequencing, miRNA has undergone complex processing processes, which may introduce corresponding deviations into the final sequencing results and directly affect the accuracy of the analysis results
[0007] In 2014, researchers such as Pieter Mestdagh wrote an article discussing that the reproducibility of miRNA sequencing results is poor, and different library construction methods may lead to significant differences in analysis results
Unable to provide real-time evaluation of sequencing data, and requires a lot of experimental resources and time
Currently there is no accepted method for establishing statistical confidence and correction for multiple testing in sequencing studies

Embodiment

[0080] This example first designed and synthesized the small RNA of known sequence used as a reference sample in the method for evaluating the quantitative results of miRNA sequencing in cancer samples, that is, the reagents used in this example for the evaluation of quantitative results of miRNA sequencing in cancer samples, and then Add exogenous known small RNA (Small RNA Spike-in mix, abbreviated as SRS) to the RNA sample, and analyze the sequencing results of SRS to judge the reliability of miRNA sequencing quantitative results. details as follows:

[0081] 1. Design of SRS

[0082] (1) Export all sequences of human miRNAs in miRBase, a public database of miRNAs, and count their length distribution and GC% distribution; wherein, the website of miRBase is http: / / www.mirbase.org / .

[0083] (2) According to the statistical results of step 1, a series of 20-22nt RNA short sequences are randomly generated, and the length distribution and GC% fall into the upper and lower quar...

Abstract

The invention discloses a reagent for miRNA sequencing quantitative result evaluation of a cancer sample and a preparation method and application thereof. The reagent comprises at least one of RNAs ofsequences shown in SEQ ID NO.1-SEQ ID NO.8. The reagent is quantitatively added into a to-be-tested RNA sample, the amount of the added reagent is quantitatively analyzed through sequencing, and a quantitative analysis result is compared with an actual added amount, thereby evaluating the accuracy and reliability of an entire miRNA sequencing quantitative result. The reagent for the miRNA sequencing quantitative result evaluation of the cancer sample and the preparation method and application thereof have the advantages that the method is simple and used easily, and the reliability of the miRNA sequencing quantitative result can be effectively evaluated; characteristics of the reagent in all aspects are very close to a real miRNA, so that the reagent can more truly, accurately and reliably reflect actual situations of a human miRNA sequencing quantitative result, thereby making more accurate and reliable evaluation to miRNA sequencing quantitative result of the cancer sample.

Description

technical field [0001] The present application relates to the field of quantitative detection of miRNA sequencing, in particular to a reagent for evaluating quantitative results of miRNA sequencing in cancer samples and its preparation method and application. Background technique [0002] Small RNA is an important class of functional molecules in organisms. Through various sequence-specific gene silencing effects, it regulates various cellular physiological processes such as cell growth and development, stress response, and silencing transposons. [0003] Small RNA sequencing is a new generation of high-throughput sequencing technology to perform high-throughput sequencing of 18-30nt small RNA in a certain species and tissue in a specific state. Then, through database comparison, the obtained small RNA sequences were analyzed and identified, and millions of small RNA sequences were classified into rRNA, tRNA, snRNA, snoRNA, and miRNA. [0004] Mature miRNAs regulate gene ex...
