Embodiment Construction

[0027] The present invention will be further described below with reference to specific embodiments. It should be understood that the following examples are only used to illustrate the present invention, but not to limit the scope of the present invention.

[0028] 1. Pretreatment and Immunomagnetic Bead Treatment

[0029] like figure 1 As shown, it is a schematic diagram of the flow pattern of the CTC detection method of the present invention. First, the peripheral blood was punctured to draw blood, and then the blood was resuspended to remove most of the red blood cells, and then CD326 magnetic beads were added for separation. After the first flow cytometry sorting, most of the lymphocytes were removed. A second flow cytometry sorting was performed to remove all lymphocytes, and finally the remaining cells were subjected to a third flow cytometry sorting to detect whether there were circulating tumor cells.

Abstract

The invention provides a circulating tumor cell detection method based on immunomagnetic beads and flow cytometry. The method comprises the following steps: performing puncture blood drawing on peripheral blood, performing blood cell re-suspending on blood, removing most of the red blood cells, and adding CD326 magnetic beads for separating; performing primary flow cytometry sorting, removing mostof the lymphocytes, performing secondary flow cytometry sorting, removing the total lymphocytes; and finally performing the third flow cytometry sorting on residual cells, namely detecting whether circulating tumor cells exist. The method disclosed by the invention is simple, easy to operate and high in detection sensitivity.

Description

technical field [0001] The invention belongs to the field of biological detection, and in particular relates to a circulating tumor cell detection method based on immunomagnetic beads and flow cytometer. Background technique [0002] Circulating tumor cells (CTCs) are the general term for various types of tumor cells present in peripheral blood. A large number of studies have shown that CTCs exist in peripheral blood in different forms, including free single CTCs and aggregated cell clusters (CTM, Circulating Tumor Microemboli). Epithelial-mesenchymal transition (EMT) occurs when tumor cells enter the peripheral blood circulation. Therefore, CTCs exist in different types, including epithelial cell phenotype, mesenchymal cell phenotype, and a mixed phenotype of epithelial and mesenchymal cells. CTM and mesenchymal phenotype CTCs have stronger metastatic potential. Early detection of CTCs in the blood plays an important role in guiding the prognosis of patients, evaluation o...
