Problems solved by technology

However, limbal stem cells form colonies as the cells expand, pushing trophoblasts away, which may lead to insufficient supply of trophoblasts
In addition, since the limbal stem cells directly contact the trophoblasts during the culture process, it is difficult to completely remove the trophoblasts during harvesting of the limbal stem cells, resulting in the risk of cross-contamination

Abstract

The invention discloses a method for culturing human limbal stem cells. The method includes the steps: pretreating a trophoblast; inoculating the trophoblast into a porous membrane; placing the porousmembrane into a container, dividing the container into a first culturing chamber and a second culturing chamber, and enabling the trophoblast to be positioned in the second culturing chamber; injecting a culture medium into the container; placing the human limbal stem cells into the first culturing chamber. Compared with learning and knowing technologies, according tot the method for culturing the human limbal stem cells, the human limbal stem cells are cultured by the aid of a culturing technique of the porous membrane, and the expansion rate of the human limbal stem cells is superior to that of human limbal stem cells cultured by a traditional culture mode. Besides, the used culture medium does not contain toxic dimethyl sulfoxide components, and the toxic killing probability of the cells is reduced.

Description

technical field [0001] The present invention relates to a method of culturing human limbal stem cells, and in particular, to a method of culturing human limbal stem cells using a porous membrane. Background technique [0002] The normality and stability of the corneal surface is extremely important to keep the cornea transparent and maintain normal physiological functions. It is not only a dam between the cornea and conjunctiva, but also a source of corneal epithelial regeneration. Corneal defects are mainly caused by trauma, bacterial infection, chemical burns and contact lenses. According to statistics, about 13 million people around the world suffer from corneal damage or disease that affects vision, and 15 to 2 million new cases are diagnosed every year. Defects in the cornea can lead to a range of eye diseases and even blindness. [0003] The discovery of Limbal Stem Cells (LSCs) is one of the most important advances in ophthalmology in recent decades. For now, trans...
