Embodiment 1

[0020] Preparation of Gardenia Water Extract

[0021] Select 100g of fresh Gardenia, soak for 30min, add 0.6L of water, and ultrasonicate twice at 80°C for 30min each time; combine the obtained filtrates, and centrifuge to obtain the supernatant at a speed of 3000 r / min; The obtained supernatant was rotatably concentrated to 100 mL under reduced pressure at 50°C to obtain a stock solution with a relative concentration of 1 g / ml; the obtained stock solution was digested at 100°C for 20 min and stored at 4°C.

Embodiment 2

[0023] In this example, gardenia water extract was used as a drug for protecting intestinal mucosal injury, and the therapeutic effect of gardenia water extract on LPS-induced intestinal mucosal injury was explored by establishing a mouse intestinal mucosal injury model.

[0024] 1. Materials and methods

[0025] (1) Drugs and reagents

[0026] LPS (E. coli O55:B5) and all other chemicals were obtained from Sigma-Aldrich (St. Louis, MO, USA).

[0027] Distilled water was filtered through the Milli-Q system of EMD Millipore. (Billerica, MA, USA);

[0028] Gardenia was purchased from Freeborn Traditional Chinese Medicine Co., Ltd. (Daqing, China) according to the regulations of the Chinese Pharmacopoeia (Chinese Pharmacopoeia Commission, 2010).

[0029] (2) Animals and Feed

[0030] ICR male mice weighing 22-25 g were purchased from the Animal Experiment Center of Harbin Medical University (Daqing) [certification number SYXK (HEI) 2014005]. The mice were controlled at a temp...

Abstract

The invention relates to a preparation method of a cape jasmine extract. The invention further relates to application of the cape jasmine extract in intestinal mucosa damage. The preparation method ofthe cape jasmine extract and the application of the cape jasmine extract in intestinal mucosa damage have the beneficial effects that by building a cape jasmine water extract model, LPS is injected respectively, then the cell factor concentrations and antioxidant indexes of duodena are measured, the histopathology situation is observed under an optical microscope, and a result shows that the capejasmine water extract can significantly inhibit LPS-induced TNF-alpha, LPS-induced IL-6 and LPS-induced IL-1; the cape jasmine water extract has anti-inflammatory and antioxidant effects by improvingtotal antioxidant capacity (T-AOC), and improving activity of total superoxide dismutase (T-SOD) and glutathione peroxidase (GSH-PX); the cape jasmine water extract has a good inhibiting effect on intestinal mucosa damage, and is a potential protective preparation of intestinal mucosa damage.

Description

technical field [0001] The invention belongs to the field of plant extracts, in particular to a preparation method and application of a gardenia extract. Background technique [0002] The gut plays a key role in digesting and absorbing nutrients, balancing the microbiome, protecting immune function, and acting as a barrier against harmful pathogens and antigens. Lipopolysaccharide (LPS / endotoxin), a major component of the outer membrane of Gram-negative bacteria, is a common cause of intestinal mucosal injury. LPS-induced cytokine release leads to pathophysiological disturbances of intestinal mucosal injury. The activation of many cellular signals is associated with LPS. LPS not only triggers immune responses but also activates pro-apoptotic signals in macrophages, endothelial cells, epithelial cells and immune cells. Despite a growing understanding of the pathophysiology of intestinal mucosal injury, its immunomodulatory mechanisms in the induction of cytokine expression...
