Problems solved by technology

[0006] Although there have been some breakthroughs in the research on CTCs, due to the scarcity (1-10 / 1mL blood) and strong heterogeneity of CTCs in the body, currently based on blood sampling (the sampling volume is only the whole body blood) 0.1%) of all the research methods are unable to obtain all CTCs in the body on the one hand, and on the other hand, due to the inability to achieve high bioactivity markers of CTCs, it is also impossible to visualize the movement trajectories and spatiotemporal heterogeneity of all CTCs in vivo
Based on the above reasons, we are still in the "blind man's touch" stage of the actual movement trajectory of CTCs in all stages of cancer, and some questions still cannot be answered, such as when are CTCs generated, which CTCs have metastasized, and which parts of the target organs have been metastasized by CTCs Location

Embodiment Construction

[0036] The present invention will be described in further detail below in conjunction with specific drawings and embodiments.

[0037] Such as figure 1 Shown is a schematic diagram of the module composition of the living animal circulating tumor cell in vitro functionalization instrument of the present invention, such as figure 2 Shown is the overall layout of the instrument for in vitro functionalization of circulating tumor cells in living animals, which includes a CTC separation module 1 , a CTC labeling module 20 , CTC reinfusion modules 15 , 16 , 17 and electromechanical control modules 18 and 19 . The CTC separation module 1 includes two upper and lower arteriovenous connecting catheters 2 and 14 with a diameter of about 1 mm, which are used to connect blood vessels of living animals to form extracorporeal circulation of blood. The arteriovenous catheter is connected to the filter device 4 through the controllable connection valves 3 and 13, wherein the filter device 4...

Abstract

The invention relates to an in-vitro functionalization instrument for circulating tumor cells of living animals. The in-vitro functionalization instrument is an integrated instrument which achieves high-capture-rate separation, high-selectivity labeling, high-activity reinfusion of the circulating tumor cells and aggregates thereof in whole blood of the living animals based on a biological micro-electromechanical system. The instrument comprises a CTC separation module, a CTC labeling module, a CTC reinfusion module and an electromechanical control module; the CTC separation module is used forseparation of all the circulating tumor cells and aggregates thereof in the whole-body blood of the small animals; the CTC labeling module is used for labeling of the circulating tumor cells; the CTCreinfusion module is used for repaid reinfusion of the visualized circulating tumor cells; the electromechanical control module is used for automatic control and signal processing of the whole instrument. The instrument can be used for visualized research on temporal and spatial heterogeneity characteristics of the CTCs and aggregates thereof in different stages of tumor generation, development, metastasis, radiotherapy / chemotherapy postoperation and the like, provides a key technical means for research on the tumor generation and development mechanism and provides a high-efficiency means foraccurate diagnosis and treatment of tumors.

Description

technical field [0001] The invention belongs to the field of biomedical engineering and relates to an in vitro functionalization instrument for living animal circulating tumor cells. Specifically, the in vitro functionalization refers to the separation, labeling and reinfusion of all CTCs in whole blood of living animals. In this way, the visualization of CTCs in living animals can be realized, and the dynamic characteristics and spatial-temporal heterogeneity of CTCs in the whole stage of tumorigenesis can be realized. Background technique [0002] According to the latest statistics from "People's Daily Online" in 2018, the incidence and mortality of cancer in my country rank first in the world. % of cancer patients die from cancer metastasis. Circulating Tumor Cell (CTC, Circulating Tumor Cell) was first proposed by Australian doctor Ashworth in 1869. It refers to the tumor that falls from the original tumor or metastases to the peripheral blood circulation due to internal...
