Problems solved by technology

[0004] The existing ECM preparation technology is quite mature, but it has some shortcomings and disadvantages, for example: (1) Most of the currently established gradient platforms are produced by diffusion, infusion, microcontact printing and microfluidic systems, etc. To study the behavior of cells and the interaction between different factors, adding too many non-biological substances; (2) The ECM biomaterials currently prepared mainly realize their specific functions by extracting a single ECM component and coating other materials; (3) The living body is a complex three-dimensional environment. After the release of cytokines, a gradient of outward diffusion is formed. The existing ECM biomaterials ignore this phenomenon and cannot accurately simulate the influence of the microenvironment in vivo on cells.

Abstract

The invention discloses a cell matrix material containing a gradient cell factor and a preparation method and application thereof. The preparation method comprises: constructing a recombinant vector of a coding gene containing a cell factor; integrating the recombinant vector to host cells to construct recombinant cells that may express the cell factor stably; subjecting the recombinant cells to medicated screening via biotin to obtain a stable transfection cell line; inoculating the stable transfection cell line to a substrate before culturing, and allowing distribution density of the recombinant cells cultured on the substrate to gradiently change in a specified direction; lyophilizing the recombinant cells attaching to the substrate to obtain the cell matrix material containing the gradient cell factor. The cell matrix material herein is capable of enriching SDF-1alpha, is originated from cells, and can minimize immunoreactions; the cell matrix material has SDF-1alpha factor gradient, so that the cell matrix material can simulate an in-vivo inflammatory micro-environment in vitro, and can also provide a support for cell attachment, so that stem cells can migrate to the inflammatory part.

Description

technical field [0001] The invention relates to a cell matrix material, in particular to a cell matrix material containing gradient cytokines, a preparation method and application thereof, and belongs to the field of biotechnology. Background technique [0002] Bio-scaffold materials are one of the hotspots in tissue engineering research. According to different sources, they are mainly divided into two categories: artificial polymer materials and natural bio-derived materials. Among them, natural biologically derived materials are derived from biological tissues. Compared with artificial polymeric materials, they have better biocompatibility and biorecognition advantages, so they have become a research hotspot in tissue engineering. Extracellular matrix (ECM) is a special natural biologically derived material, which is prepared by removing the cellular components in the prototype tissue of the organism through special physical and chemical methods. Recent studies have shown...
