Abstract

The invention relates to the technical field of tissue culture, in particular to a directional culture medium of germinating seeds of oreorchis patens(Lindl.)Lindl. and a directional culture and seedling method. The directional culture medium is prepared from a proliferation culture medium which is an MS culture medium containing sucrose, agar powder, 6-BA and NAA, a differentiation culture mediumwhich is an MS culture medium containing sucrose, agar powder, KT and IBA and a rooting culture medium which is an MS culture medium containing sucrose, agar powder, 6-BA and IAA. Accordingly, the germinating seeds of oreorchis patens(Lindl.)Lindl. serve as experimental materials, the regulation and promotion effects of different mitogens and auxins and ratios on the development process from thegerminating seeds to seedlings are studied, the results show that the 6-BA and the NAA are beneficial to protocorm proliferation, the KT and the IBA are beneficial to differentiation of protocorm intocorm, and the 6-BA and the IAA are beneficial to corm rooting. According to the method, the germinating seeds of oreorchis patens(Lindl.)Lindl. grow directionally, the seedling time is shortened, andthe seedling efficiency is improved.
