Embodiment 1

[0060] Please refer to figure 1 , the embodiment of the present invention provides a method for detecting α-amino nitrogen in a tobacco leaf sample by a continuous flow method, comprising the following steps:

[0061] Moisture was determined by oven method (YC / T 31), and the content of α-amino nitrogen in different tobacco leaf samples was determined by continuous flow analysis.

[0062] 1) Sample pretreatment:

[0063] Tobacco leaf samples were ground and passed through a 0.45 mm sieve (40 mesh), and their moisture content was determined. Accurately weigh about 0.5g of tobacco leaf sample (accurate to 0.1mg), put it into a 100mL dry Erlenmeyer flask with a stopper, add 50ml of 5% acetic acid solution, cover the stopper, extract by mechanical vibration at room temperature for 30min, and take the extract after standing still. The solution was filtered with Whatman No.40 filter paper, and the filtrate was collected for analysis.

Embodiment 2

[0093] The embodiment of the present invention provides a method for detecting α-amino nitrogen in cut tobacco samples of finished cigarettes by continuous flow method, comprising the following steps:

[0094] 1) Sample pretreatment:

[0095] The shredded tobacco was stripped from the finished cigarette sample, dried at 40°C, ground and passed through a 0.45mm sieve (40 mesh), and its moisture content was measured. Accurately weigh about 0.5g of the sample (accurate to 0.1mg), place it in a dry 100mL Erlenmeyer flask with a stopper, and add 50ml of 5% acetic acid solution. Cover the stopper, extract by mechanical shaking at room temperature for 30 min, take the extract and filter it with Whatman No. 40 filter paper after standing still, and collect the filtrate for analysis.

[0096] 2) The preparation of reaction reagent and working standard solution is the same as in Example 1.

[0097] 3) Determination of actual samples:

Abstract

The invention relates to the technical field of tobaccos and particularly relates to a detection method for determining alpha-amino nitrogen in tobacco or a tobacco product by using a continuous flowmethod. The detection method comprises steps of reacting ninhydrin with alpha-amino nitrogen of tobacco or the tobacco product under the condition that a pH value is 5.4-5.6 to generate a stable compound, measuring the absorbance of the stable compound at 570nm, and calculating the concentration of the alpha-amino nitrogen in extract liquor of the tobacco or the tobacco product through a predetermined standard curve. The detection method is advantaged in that a blank that no detection method for determining alpha-amino nitrogen in the tobacco or the tobacco product by a continuous flow methodexists at present can be filled.

Description

technical field [0001] The invention relates to the technical field of tobacco, in particular to a detection method for measuring α-amino nitrogen in tobacco or tobacco products by a continuous flow method. Background technique [0002] Tobacco chemistry research has carried out qualitative analysis on thousands of compounds in tobacco leaves and smoke, and there are more and more requirements for quantitative analysis of chemical components in tobacco and tobacco products, including the update and improvement of analytical methods , the improvement of analysis efficiency, and the gradual modernization of analysis methods. This is an important basic work for the stability and development of cigarettes. The chemical composition of tobacco or tobacco products has a great influence on the quality of tobacco leaves and the sensory quality of cigarettes. At present, the quantitative analysis of conventional chemical indicators in tobacco or tobacco products includes total sugar...
