Problems solved by technology

However, chemotherapy drugs cannot specifically kill cancer cells, and some normal cells can also be killed, such as bone marrow, intestinal mucosa and hair follicles
In addition, some cancer cells can survive chemotherapy and become resistant to chemotherapy drugs. Some chemotherapy drugs can even stimulate normal cells to mutate into cancer cells. Cancer stem cells are not easily affected by chemotherapy drugs. After a period of treatment, cancer Stem cells can quickly cause cancer to recur

Embodiment 1

[0123] Example 1 Galectin-3 expression and upstream miRNA prediction in pancreatic cancer cell lines

[0124] 1. Use Western Blot and qRT-PCR to confirm the expression of Galectin-3 in normal pancreatic ductal cells and pancreatic cancer cell lines. Western Blot and qRT-PCR were used to detect the expression of Galectin-3 in normal pancreatic ductal cells and pancreatic cancer cell lines.

[0125] Bioinformatic analysis of the relationship between Galectin-3 and pancreatic cancer using the GEPIA platform

[0126] It was found in the platform that by comparing the expression of Galectin-3 in 179 cases of pancreatic cancer specimens and 171 cases of normal specimens, the statistical analysis found that the expression of Galectin-3 in pancreatic cancer was significantly higher than that in normal people ( figure 1 A). Moreover, among pancreatic cancer patients, the survival time of the Galectin-3 low expression group (89 cases) was longer than that of the Galectin-3 high ex...

Abstract

The invention discloses miRNA-128-3p of targeting a Galectin-3 gene. A sense strand of the miRNA-128-3p is 5'-CACUAGAUUGUGAGCUCCUGGA-3', and an antisense strand of the miRNA-128-3p is 5'-UCCAGGAGCUCACAAUCUAGUG-3'. The invention further discloses a drug carrier for the miRNA-128-3p of targeting the Galectin-3 gene, and the drug carrier is an exosome (EXO) derived from human umbilical cord mesenchymal stem cells (HUCMSCs). In addition, the invention further discloses experimental verification of use of the miRNA-128-3p, carried by the HUCMSCs exosome, of targeting the Galectin-3 gene to resistance of the pancreatic cancer. According to the embodiments of the invention, miRNA-128-3p mimic is successfully transfected to a neonatal umbilical cord tissue-derived HUCMSCs-EXO to construct an engineered EXO; and the engineered EXO can inhibit migration, invasion and proliferation of pancreatic cancer cells by targeting Galectin-3.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and specifically relates to a HUCMSCs exosome targeting Galectin-3 gene carrying miRNA-128-3p and its anti-pancreatic cancer application. Background technique [0002] Pancreatic cancer is a common malignant tumor of the digestive system in clinical practice. The mortality rate within 1 year after diagnosis is about 75%, and the 5-year survival rate is less than 6%. In European and American countries, pancreatic cancer is the fourth most common malignant tumor. In my country, the annual incidence of pancreatic cancer is about 511 / 100,000, and it is estimated that 50,000 to 60,000 new cases will be added each year. Most patients with pancreatic cancer have no obvious discomfort in the early stage, and they are already in the advanced stage when they are discovered. They miss the chance of radical surgical resection and can only rely on radiotherapy and chemotherapy. However, chemotherapy dr...
