Problems solved by technology

It has been reported that the downregulation of mitochondrial oxidative phosphorylation can cause premature aging in somatic cells and mice; more interestingly, many researchers have recently found that mitochondria and energy metabolism in adult / embryonic stem cells (ESCs) The function of normal stem cells may affect the proliferation, differentiation potential and survival period of normal stem cells, etc.

Embodiment 1

[0019] 1. Experimental Methods and Materials

[0020] The UL16 expression plasmid pET-31b(+)-UL16 of HSV-1, the expression vector pET-31b(+) was purchased from Invitrogen Company, and the plasmid pFLAG-CMV-2 was purchased from Sigma Company. Herpes simplex virus F strain (HSV-1 F), strain DH5α receptor bacteria and cells are preserved in our laboratory.

[0021] Primer design and synthesis

[0022] According to the sequence of HSV-1 UL16 gene queried on NCBI, primers UL16-for and UL16-rev were designed by Primers5.0 software for the amplification of HSV-1 UL16, and the primers were synthesized by Shanghai Sangong Company.

Abstract

The invention discloses application of UL16 protein in preparation of a small molecular medicine for promoting in-vitro differentiation of embryonic stem cells, and application of stem cell transplantation in treatment of nervous system degenerative diseases. According to the invention, by construction of UL16 plasmids to transfect host cells, the fact that UL16 can promote aerobic oxidation and oxidative phosphorylation of glucose and activate the cell productivity pathway so as to increase the intracellular ATP content and improve mitochondrial functions is found. Finally, by construction ofUL16 expression plasmids to transfect mouse embryonic stem cells, mitochondrial function state enhancement and energy metabolism enhancement are found, and stem cell differentiation can be promoted.Clinical treatment of cells in a differentiation defect state and cells with a low differentiation rate is realized; and the UL16 protein is used for treating related diseases or pathological and physiological processes of mitochondrial dysfunction or hypofunction of the cells.

Description

technical field [0001] The field of protein screening and application mechanism of the present invention, more specifically, the present invention relates to a protein UL16 that has the function of enhancing the functional state of mitochondria and increasing the differentiation efficiency of stem cells and its application. Background technique [0002] Embryonic stem cells (ESCs) provide seed cell sources for the cell replacement therapy of various metabolic liver diseases due to their ability of self-renewal, high proliferation and multi-directional differentiation, making them a research hotspot, but their induction The low efficiency of differentiation has been a key problem that is difficult to solve. In vitro, mouse cells can be directed to differentiate into almost all types of adult cells; human cells can also be successfully differentiated into neurons, liver cells, endothelial cells, cardiomyocytes, pancreatic cells and hematopoietic cells and other types of cell...
