Embodiment 1
[0032] Example 1 Nobiletin alleviates the effect of 5-fluorouracil on the proliferation toxicity of HaCaT cells
[0033] The cultured skin cells HaCaT were digested, counted, seeded into a 96-well plate, and 5,000-10,000 cells were planted in each well. After culturing for 24 hours, each well was divided into a blank solvent control group, a 5-FU group, and a 5-FU+nobietin group. final concentration as figure 1 shown. After continuing to culture for 48 hours, the Cell Counting Kit-8 (CCK-8) detection kit (C0037, purchased from Shanghai Biyuntian Biotechnology Co., Ltd.) was used to measure the survival rate of the cells, so as to calculate the proliferation toxicity of 5-FU to the cells and the Sichuan Alleviating effect of nobiletin on proliferative toxicity. GraphPad Prism 6.0 software and t test were used for statistical analysis and drawing of the results. figure 1 Experimental results are listed.
Embodiment 2
[0034] Example 2 Nobiletin can alleviate the hand-foot syndrome caused by capecitabine on the rat model
[0035] Rat animal model was constructed. Capecitabine was given to 6-week-old female SD rats by intragastric administration every day. After a few days, hand-foot syndrome appeared in the paws of the rats (photograph as figure 2 shown). There is no difference between the left and right paws in the hands and feet, and the degree of hand-foot syndrome in the two paws is similar. Similar to humans, rats developed hand-foot syndrome in the paws after oral administration of capecitabine. The etiology of the two is exactly the same, and the symptoms are very similar. Therefore, rats are very good animal models for simulating capecitabine-induced hand-foot syndrome.
[0036] After the SD rats were reared and adapted for one week (about 200 g), the rats were divided into 12 in each group, and then the oral administration test was carried out. Capecitabine was dissolved in ca...
