Problems solved by technology

[0004] Because low temperature can inhibit the growth of bacteria, some researchers have proposed low temperature storage of platelets. However, since platelets are living cells, deep low temperature (-80°C) preservation of platelets can prolong the storage period, and clinical application has proved its immediate hemostatic effect Obviously, but the disadvantages are: 1) It needs a very large and expensive cryogenic storage refrigerator (-80 ℃) or consumes a lot of liquid nitrogen; 2) It is inconvenient to carry; 3) It is difficult to transport over long distances; especially 4) After freezing Platelets can fatally impair their biological activity

Embodiment Construction

[0038] The invention will be further described below in connection with the specific embodiments, but the embodiments are merely exemplary, and the scope of the invention is not limited. It will be understood by those skilled in the art that the details and forms of the technical solutions of the present invention can be modified or replaced without departing from the spirit and scope of the invention, but these modifications and replacements fall into the scope of the invention.

[0039] First, platelets best preservation temperature selection

[0040] The present invention provides an extension of platelet preservation, from platelet apoptosis, aggregation function, thrombosis capability, and in vivo platelet clearance rate, to select the optimal temperature of the platelet. The four indicators of JC1, PS, CD62P, and PAC1 were selected to detect the apoptosis activation state of platelets. JC1 detects film potential depolarization, which is a specific indicator of the apoptosis ...

Abstract

The invention discloses a platelet preservation method. According to the invention, the influence of different temperatures on the stored platelets is discussed from the aspects of apoptosis and activation of the stored platelets, loss of physiological functions of the platelets, hemostasis thrombus formation ability of the platelets returned to the body and removal efficiency, and finally, the optimal temperature for storing the platelets is selected and is higher than 4 DEG C and not higher than 8 DEG C, preferably 6 DEG C; the platelets stored at the temperature have the best physiological function, the best thrombosis ability and the slowest in-vivo clearing efficiency. Compared with the existing accepted preservation temperature of 4 DEG C and 22 DEG C, the preservation period of the platelets is prolonged, the physiological functions of the platelets are kept to the maximum extent, meanwhile, the requirements of simple and convenient platelet storage method, long-term mass preservation, convenient transportation and the like are met, and the method is suitable for clinical general popularization and application. Meanwhile, the mechanism of apoptosis of the platelets at different preservation temperatures is also researched, and the reasons of apoptosis caused by preservation of the platelets at low temperatures (4 DEG C and 6 DEG C) and 22 DEG C are obtained.

Description

Technical field [0001] The present invention relates to a preservation method, and more particularly to a platelet preservation method. Background technique [0002] Platelet is a non-nucleated ingredient from mature mega-cytoplasm that plays a crucial effect in hemostasis and thrombosis, which is widely used to treat hemorrhagic diseases caused by platelet reduction and platelet dysfunction. And patients with bone marrow inhibitory chemotherapy. With the development of medical technology, its clinical application is increasing, but platelet products are still in a case where severe supply is short-term, and future supply and demand contradictions will be more prominent. Therefore, platelet transfusion protection is still the frontier problem of the world. Due to the short bloody life, structural and function is susceptible to a variety of factors, the preservation of platelets is urgently needed to achieve the following goals: the storage method is simple, long-term preservation...
