[0027] 1. Concanavalin A-induced immune liver injury model in mice

[0028] The mouse model of liver injury caused by tail vein injection of concanavalin A is a recognized classic animal model for simulating human autoimmune hepatitis. Balb / c mice, male, weighing 18-22 g. Food and drink were not restricted during feeding, and a 12-h day / night rhythm was maintained. Balb / c mice were randomly divided into 4 groups according to body weight, 15 in each group. The four groups were control group, concanavalin-induced immune liver injury model group, isosteviol 50 mg / kg dose treatment group and isosteviol 100 mg / kg dose treatment group. The mice in the control group were injected with sterile normal saline into the tail vein; the mice in the model group and the mice in the isosteviol treatment group were injected with concanavalin A (Con A) normal saline solution at a dose of 20 mg / kg. Subsequently, the mice in the model group were gavaged with normal sali...

Abstract

The invention discloses an application of isosteviol in preparation of a medicine for treating autoimmune hepatitis. Compared with the prior art, the invention discovers that the isosteviol can be used for improving the immune liver injury of mice induced by concanavalin A (concanavalin A). The isosteviol can relieve mouse liver cell swelling and necrosis induced by concanavalin A. The isosteviol is used for inhibiting the rise of serum glutamic-pyruvic transaminase, glutamic oxalacetic transaminase and lactic dehydrogenase levels; the levels of serum inflammatory factor interleukin 6 (IL-6), tumor necrosis factor alpha (TNF-alpha) and interleukin 1 beta (IL-1beta) are reduced. The expression of IL-6, TNF-alpha, IL-1beta, macrophage inflammatory protein 1alpha, intercellular adhesion molecule 1 and C-X-C motif chemotactic factor 10 in the liver tissue of the mouse is reduced. The effects show that the isosteviol can be used for treating the autoimmune hepatitis and has the prospect of developing medicines.

Description

technical field [0001] The invention relates to the field of new applications of isosteviol, in particular to the application of isosteviol in the preparation of medicines for treating autoimmune hepatitis. Background technique [0002] Autoimmune hepatitis (AIH) is an inflammatory disease of the liver caused by abnormal immune response to liver tissue. Is a T cell-mediated autoimmune response in genetically susceptible individuals. Similar to all other autoimmune diseases, the exact pathogenesis of AIH is unknown and may be related to genetic, environmental and accidental factors. Several HLA genotypes encoding major histocompatibility complex proteins increase the risk of developing AIH. AIH can manifest from very mild subclinical disease to fulminant acute liver failure, and its clinical features include elevated serum aminotransferase levels, hyperimmunoglobulin Gemia, positive serum autoantibodies, and moderate to severe liver histology interface hepatitis, etc. AIH...
