The present invention relates to biomedicine, and is especially the method of inducing stem cell to differentiate into pancreatic island-like cell and the application of pancreatic island-like cell. The present invention aims at providing extracorporeal method to obtain great amount of pancreatic island-like cell and applying inducing differentiated pancreatic island-like cell as seed cell for transplanting pancreatic island cell, preparing microcapsule stem cell preparation and acting as pharmacokinetics model in cell medicine screening. The technological scheme of the present invention includes: 1. obtaining GLP-1R gene; 2. constructing virus carrier containing GLP-1R gene; 3. separating, culturing and amplifying stem cell; 4. adding cell factor GLP-1 to the culture liquid to induce stem cell to differentiate into pancreatic island-like cell; and 5. identifying the obtained pancreatic island-like cell. The present invention has excellent application foreground in regeneration medicine.
Description
Method for inducing stem cells to differentiate into islet-like cells and application of islet-like cells technical field The invention relates to the field of biomedicine, in particular to a method for inducing stem cells to differentiate into islet-like cells in vitro and the application of the islet-like cells. Background technique Diabetes mellitus (DM, Diabetes Mellitus) is a disorder of sugar, fat and protein metabolism caused by relative or absolute deficiency of insulin secretion or insulin receptor deficiency. Clinically, type I and some type II diabetes are mostly treated with subcutaneous injection of insulin. Cell therapy is an ideal treatment strategy for diabetes, especially type I diabetes. For diabetic patients who have lost the function of islet cells, implanting insulin-secreting β cells or their substitutes is the most ideal treatment method. In recent years, pancreatic islet cell transplantation has achieved some curative effects in the treatment of di...
