The ratio of OD260 to OD280 of the extracted total RNA of the sandalwood wood tissue is 1.8 to 2.0; the ratio of OD260 to OD230 is 2.0 to 2.3; the concentration of the RNA water solution is 350.0-500.0ng/mu L; electrophoresis detection shows that the RNA strip is clear and good in integrity; and the brightness at 28S is 1.9-2.1 times that at 18S. The total RNA of the sandalwood wood tissue extracted by the method provides a good foundation for related researches of sandalwood molecular biology, and the method has relatively high popularization and application values.

The difference between SPS gene expression quantities of the test materials in different growing periods are quantitatively analyzed, and the relative concentration of the SPS gene expression and the sucrose content are subjected to relevance analysis; a result shows that the relative concentration of the SPS gene expression and the sucrose content are obviously relevant; and a quick detection method for the sweet sorghum SPS gene is established according to the result of the invention, so as to achieve the aims of quickly detecting and determining the best harvest time and screening novel sweet sorghum materials with high sucrose content.
