Axenic germination method of roegneria kamoji plants

A technology of goose grass and plants, which is applied in the field of aseptic germination and cultivation of the seeds of the genus Gooseberry, which can solve the problems of small seeds, incomplete disinfection of seeds, and non-germination of seeds of the genus Gooseberry, and achieve the goal of overcoming germination. Inconsistency problem, shortened disinfection time, and the effect of overcoming the phenomenon of weak seedlings

Problems solved by technology

[0003] Under the background of this research, the inventors found that the seeds of the genus Esperanza have small grains, less storage of nutrients, high dormancy, and inconsistent germination. There are many difficulties in obtaining neat and consistent aseptic seedlings.
At present, the aseptic seeding and germination technology has been successfully applied to rhizome plants such as flowers and medicinal materials. If the existing technology is applied to the plants of the genus Aphidia, there are many deficiencies: first, due to the dense structure of the seed coat of the genus A. One of the difficulties is disinfection. According to existing methods such as the patented orchid plant seed aseptic germination and plant cultivation method, patent number: 200510010764; soybean transgenic neutron disinfection treatment method, patent number: 02150780.5, use the above disinfection method None of the methods are suitable for the plants of the genus Escherichia genus. Either the seed disinfection is not thorough, a large amount of materials are contaminated, or the disinfectant treatment time is too long, which will cause poisonous effects on the seeds and cause deformed seedlings; secondly, it cannot solve the problem of inconsistent germination. It was found that many of the seeds inserted into the medium did not germinate, and uniform sterile seedlings could not be obtained.

Embodiment 1

[0017] Source of material: Collected from Qingshuihe County, Hohhot City, Inner Mongolia, Eguangrass 05-68 material, cultivated in resource nursery, and harvested when the seeds are ripe.

[0018] Training method:

[0019] 1. Seed pretreatment: choose plump seeds that are not affected by diseases and insect pests, wash them with tap water for 15-20 minutes, place them at 4°C and soak them in sterile water for 24 hours;

[0020] 2. Disinfection of explants: Peel off the seed coat of the above-mentioned pretreated seeds, first disinfect with 75% alcohol for 30 seconds, wash with sterile water for 3 times, then disinfect with 0.1% mercuric chloride solution for 3 minutes, and wash with sterile water 5 times, and finally use sterile filter paper to blot the water on the surface of the seeds, and inoculate them into solid medium; medium: MS medium, inoculate the seed embryos upward;

[0021] 3. Culture conditions: During the entire culture process, the aseptic germination of the s...

Embodiment 2

[0024] Source of material: Collected from Pianguan, Shanxi Province, the genus 05-28 of the genus Esperus, cultivated in the resource nursery, and harvested when the seeds are ripe.

[0025] The difference with Example 1 is that in step 1, select plump seeds that are not affected by diseases and insect pests, rinse them with tap water for 15 minutes and place them at 4°C to soak them in sterile water for 12 hours; Embryos were inoculated upwards.

Embodiment 3

[0027] Source of material: Collected from the Aershan Haosengou Forest Farm, the 05-11 material of the genus Esperia genus, cultivated in the resource nursery, and the seeds are harvested at the ripening stage.

[0028] The difference from Example 1 is that in step 1, seeds that are plump and not affected by diseases and insect pests are selected, rinsed with tap water for 15 minutes, and then placed at 4° C. and soaked in sterile water for 20 hours.

Abstract

The invention belongs to the field of biotechnologies, in particular to an axenic germination and culture method of roegneria kamoji plant seeds. The method comprises the following steps: seed preprocessing: selecting plump seeds which are not damaged by diseases and insects, washing the seeds by tap water for 15-20min, and steeping the seeds in sterile water at 2-4 DEG C for 12-30h; then explantsterilizing: peeling off the flower glume of the preprocessed seeds, sterilizing by alcohol with the alcoholic strength of 75 percent for 25-40s, washing by sterile water for 3 times, sterilizing by 0.1 percent of mercuric chloride solution for 1-4min, washing by sterile water for 5 times again, then sucking the moisture on the surfaces of the seeds by sterile filter paper, and then transferring the seeds to a solid culture medium which is an MS;1 / 2MS culture medium; firstly, culturing in a light-proof enclosure, then adopting lighting culture when buds grow to be 1-2 cm in height, and then culturing sound seedlings, namely, the seedlings are transferred to a 1 / 2MS culture medium when the seedlings grows to be 8-10 cm in height so that the height of the seedlings is kept to be 10+ / -2 cm. The invention improves the germination ability of roegneria kamoji plants, increases the germination index from 14.48 to 42.06 and obtains uniform sterile seedlings, thereby satisfying the requirements of seed planting and genetic breeding experiments of roegneria kamoji plants.

Description

technical field [0001] The invention belongs to the field of biotechnology, and in particular relates to a method for aseptic germination and cultivation of the seeds of the genus Amaranthus. Background technique [0002] The genus Roegneria was established by C. Koch in 1848 with R. caucasica C. Koch as the model species. It is the largest genus in the Triticea family. There are more than 130 species in the world. The genus Escherichia is a kind of plant with warm and cold characteristics. It is widely distributed and concentrated in Asia, Europe and North America. Already on the verge of the coast of the Arctic Ocean, the southern boundary extends to my country's Yunnan, Guangxi, and Taiwan regions and the Rocky Mountains in the western United States. There are more than 70 species of the genus Escherichia in my country, which are distributed from the Turpan Basin at -30 meters below sea level to the Himalayan ridge at an altitude of 5,500 meters (Journal of Northwest Bot...
