Problems solved by technology

[0003] At present, Qigang adopts rhizomes and divisions to propagate, and each clump can only multiply by 3-4 times, which cannot meet market needs.
Cutting is a simple and widely used vegetative propagation technique in production, but the stems of gramineous herbs are hollow, and cutting is not easy to succeed. So far, there have been no reports of successful propagation by cuttings at home and abroad.

Embodiment 1

[0024] On September 6, 2009, two-year-old Qigang was cut in the Xiaotangshan Base of Beijing Grass Industry and Environmental Research and Development Center, and healthy stems with a stem diameter of 1-1.5cm and axillary buds were selected as the source of cuttings. Cut the stalks into 10-12cm long sections as cuttings, leave 1 node for each section, leave 2-3cm below the node, cut into oblique openings, leave 7-8cm above the nodes, and cut into flat openings. Soak in water, NAA and IBA to accelerate germination, and soak for 24 hours. 15mg·L for cuttings on July 13 -1 NAA soaking treatment. The cutting substrate is vermiculite, and the vermiculite should be poured thoroughly before cutting, and the cutting environment should be kept moist during the experiment. The cutting bed is covered with plastic film to keep moisture. The daily temperature / night temperature of the substrate is controlled to 30-35°C / 15-20°C through geothermal lines, and the relative air humidity is 60...

Embodiment 2

[0026] On September 15, 2009, stem cuttings in different parts were carried out. Stem 9 joints are cut into top, middle and base 3 sections, and the cutting method of each cutting is the same as embodiment 2. The germination rate of the axillary buds of Qigang stems was 62.1% when the top stems were potted. After 20 days of rooting induction, the rooting rate of one-time seedling investigation was 92.5%. The germination rate of the axillary buds of Qigang stems was 92.3% when the middle stem was potted. After 20 days of rooting induction, the rooting rate of one-time seedling investigation was 92.5%. The germination rate of the axillary buds of Qigang stems was 100% when the base stems were potted. After 20 days of rooting induction, the rooting rate of the plants was 100% in one-time seedling investigation.

Embodiment 3

[0028] The cutting procedures were all carried out as in Example 1, except that the day temperature / night temperature for axillary bud induction was 35-38°C / 15-20°C. The germination rate of Qigang stem axillary buds was 93.4%. After 20 days of rooting induction, the rooting rate of the plants was 100% in one-time seedling investigation.

Abstract

The invention provides a cutting propagation method of Miscanthus giganteus, which comprises the steps of: (1) cutting treatment of a mature stem of the Miscanthus giganteus: stripping the stem from a leaf sheath and cutting into segments of 10-12cm as shoots for cutting, and obliquely cutting into a substrate after induced hormone treatment of axillary bud; (2) germination and cultivation of the axillary bud: controlling the temperature of the substrate at high temperature through warming geothermal lines for inducing the axillary bud to germinate and come out of the soil, and cultivating the growth of the bud by controlling the day temperature / night temperature at 20-30 DEG C / 15-20 DEG C respectively; and (3) rooting cultivation of the bud: transplanting the bud to a nutrition pot after the bud grows to 15-20 cm or more, paving the geothermal lines to control the temperature of the substrate and promoting the rooting of the bud. The method has the advantages of high propagation efficiency, robust cuttage root system and high transplanting survival rate, and is a mature technology suitable for commercial propagation of the Miscanthus giganteus.

Description

technical field [0001] The invention relates to the cutting propagation technology of Miscanthus×giganteus, in particular to utilize the mature stalks of the Miscanthus giganteus to promote the generation of axillary buds and rooting. Through this propagation technology, the problem of low reproduction efficiency of Miscanthus using ramets can be solved. Each cluster of Qigang (15 stems) can obtain 100-120 cutting seedlings, which can be multiplied by more than 100 times, and the reproduction efficiency is high. Large-scale and commercial production of Qigang seedlings. Belongs to the field of biotechnology. Background technique [0002] Qigang is a perennial herbaceous plant in the Poaceae family. It is light-loving, cold-resistant, early-dawn-resistant, waterlogging-tolerant, and has strong stress resistance. It has great application potential in garden landscape, road greening, windbreak and sand fixation, and can also be used for feed, building materials, papermaking, ...
