Embodiment Construction

[0050] Below in conjunction with accompanying drawing, take lotus leaf extract as example, the present invention is described in further detail. Except lotus leaf extract, the present invention is also suitable for screening triglyceride inhibitors from other plant extracts.

[0051] 1. Preparation of hollow fiber for adsorbing triglyceridease

[0052] 1) Reagent and solution preparation:

[0053] Porcine pancreas triglyceride enzyme (Simga company);

[0054] Polypropylene hollow fiber, specifications: inner diameter 335 μm, wall thickness 100 μm, pore 0.2 μm (Hangzhou Hengfilter Membrane Technology Engineering Co., Ltd.);

[0055] Bradford protein concentration determination kit (Beiyuntian Company).

[0056] 2) Preparation of hollow fiber for adsorbing triglyceridease

[0057] Soak 50 polypropylene hollow fibers with a length of 6 cm in 100% ethanol and activate them for 1 hour, then take them out, clean the attached ethanol with deionized water; add 10 mL (0.1 mg / mL) tr...

Abstract

The invention discloses a method for screening a triglyceride enzyme inhibitor from a plant extract. The method comprises the following steps of: performing enzyme immobilization by utilizing an adsorption carrier; performing affinity selective separation by utilizing the adsorption carrier loaded with an enzyme to obtain an active compound from the plant extract. According to the method disclosed by the invention, a hollow fiber is used as the adsorption carrier, and a traditional active screening mode of screening after separating is improved into a novel active screening mode of separating after screening and identifying, and thereby, the blindness of compound preparation and separation is greatly reduced. The method can be popularized to be applied to rapidly screen compounds with triglyceride enzyme inhibitory activity in natural products or traditional Chinese medicine mixtures.

Description

technical field [0001] The invention belongs to the field of drug screening and evaluation methods, and relates to a method for screening triglyceride inhibitors from plant extracts. Background technique [0002] Triglycerides are the most abundant lipids in the human body. Triglycerides in adipose tissue are decomposed into glycerol and fatty acids under the action of a series of lipases. This process is called lipid mobilization. Fatty acids generated by lipid mobilization can be released into the blood, combined with albumin to form fatty acid albumin, and transported to other tissues for utilization; glycerol is transported to the liver, catalyzed by glycerol kinase to generate 3-phosphoglycerol, and enters the glycolysis pathway for decomposition or use In gluconeogenesis. [0003] During lipid mobilization, triglycerides, which catalyze the hydrolysis of triglycerides to diglycerides, are the rate-limiting enzymes of lipid mobilization. If a high-efficiency inhibitor...
