Problems solved by technology

However, high concentration of HCG in some samples can lead to a high level of "hook effect", that is, when the concentration of HCG is far greater than the detection limit of colloidal gold test strips, the measured results of HCG will decrease with the increase of concentration. The color of the T line becomes lighter instead, causing false impressions of low levels of HCG in clinical diagnosis, making the diagnostic results untrue

[0026] Preparation and detection application of embodiment 1HCG detection test strip

[0027](1) Preparation of colloidal gold-labeled mouse anti-HCG-β monoclonal antibody marker pad

[0028] A colloidal gold solution with a diameter of 40nm was prepared by the chloroauric acid-trisodium citrate reduction method. After the preparation was completed, three parts of the colloidal gold solution were taken, respectively, with 0.2MK 2 CO 3 Adjust the pH of the solution to pH7.5, pH8.5, pH9.5, then place the solution on a magnetic stirrer and stir slowly, and slowly add 0.5mg, 1.0mg, 1.5mg of mouse anti-HCG-β monoclonal antibody per 100ml solution Add it dropwise into the colloidal gold solution, continue to stir for 1 hour, then add 0.2% BSA and 0.1% polyethylene glycol 20000 at a final concentration to block for 30 minutes, centrifuge at 12000 rpm for 30 minutes, discard the supernatant, and reconstitute with the colloidal gold working solution. Dissolve to 76.5ml, spread 16cm a...

Abstract

The invention relates to a test strip and a method for fast quantitative detection of human chorionic gonadotropin (HCG). The test strip comprises a sample loading pad, a marker pad, an NC membrane, a sample suction pad and a plastic plate. The sample loading pad, the marker pad, the NC membrane and the sample suction pad are orderly adhered to the plastic plate. The marker pad is coated with colloidal gold-marked mouse anti-human HCG-beta monoclonal antibodies. The NC membrane is coated with a detection line T composed of mouse anti-human HCG-alpha antibodies and a quality control line C composed of goat anti-mouse IgG antibodies. On the NC membrane, a detection line T side close to the marker pad is provided with a sample loading indication line for indicating a sample loading position. Through loading of a sample on the NC membrane, the test strip can solve the problem that the existing detection reagent produces a hook effect because of too high HCG content, and can realize quantitative detection of HCG in blood or urine by a matched immunochromatographic assay interpretoscope.

Description

technical field [0001] The invention relates to the technical field of biological applications, in particular to a test strip and a detection method for rapid quantitative detection of HCG. Background technique [0002] When the fertilized egg is implanted in the uterus, the pregnant woman will secrete a new hormone called human chorionic gonadotropin (Human Chorionic Gonadotrophin, referred to as HCG). HCG was first discovered in the urine of pregnant women by Aschheim and zondek in 1927. HCG is a hormone secreted by cytotrophoblast and syncytiotrophoblast. It first exists in the placenta and is excreted into urine through blood circulation. HCG and its metabolites are present in the placenta, blood and urine. About 10 days after conception, HCG can be detected in the morning urine of pregnant women. In the serum of normal pregnant women, the concentration of HCG begins to increase 7 to 10 days after the corpus luteum reaches its peak or 4 to 7 days after the fertilized ...
