The method comprises the following steps: extracting the ganoderma lucidum spore oil from wall-broken ganoderma lucidum spore powder; adding organic solvents such as petroleum ether and ethanol and purified water to extract; concentrating extracted lower-layer liquor; carrying out column separation on the concentrated liquor by using a macroporous resin column; carrying out gradient eluting by purified water, 50% ethanol and ethanol with concentration greater than or equal to 75%; and collecting the last eluate and freeze-drying the eluate to obtain the anti-tumor composition. The composition obtained by the invention can inhibit external and internal tumor cell growth, and the anti-tumor rate reaches over 56.3%.
