Embodiment 1

[0065] Example 1: Application of adipose-derived mesenchymal stem cells (AMSC-miR-122) carrying miR-122 in chemosensitization of liver cancer ectopic tumor-bearing models.

[0066] 1. Preparation of adipose-derived mesenchymal stem cells (AMSC):

[0067] Adult adipose tissue collected by liposuction was washed with DPBS, digested with 0.075% type Ⅰ collagenase (Sigma) at 37°C for 1 hour, then centrifuged at room temperature, discarded the upper layer of fat and liquid, and the cell pellet was resuspended in DPBS and washed twice Afterwards, the cells were collected by centrifugation, inoculated in a plastic culture bottle of low-sugar DMEM medium (Gibco) containing 5% fetal bovine serum (MSC is suitable for fetal bovine serum), and the medium was changed after 24 hours to remove blood cells. Adherent cells depended on growth 2-3 days and a half amount of medium was changed, and when the proliferation was close to 80% confluence, digestion and passage were carried out, and seve...

Embodiment 2

[0100] Example 2: Application of adipose-derived mesenchymal stem cells (AMSC-miR-122) carrying miR-122 in chemotherapy sensitization of an orthotopic tumor-bearing model of liver cancer.

[0101] 1. The construction and verification methods of AMSC-miR-122 are the same as before;

[0102] 2. Establishment of an orthotopic tumor-bearing model of liver cancer:

[0103] Six-week-old nude mice were anesthetized with pentobarbital sodium, and inoculated 2×10 6 HepG2-luci liver cancer cells were used to detect the bioluminescent signal of luciferase in real time by small animal in vivo imaging technology every week for 5 consecutive weeks to confirm the establishment of a tumor-bearing nude mouse model of liver cancer.

[0104] 3. AMSC-miR-122 infusion: AMSC-miR-122 was diluted with DPBS and infused by tail vein;

[0105] The tail vein infusion of AMSC-miR-122 is as follows: AMSC-miR-122 is diluted to 1×10 by DPBS 7 / mL concentration, the effective dose of tail vein infusion is ...

Abstract

The invention belongs to the field of stem cells and biotechnology, and relates to a novel application of adipose-derived mesenchymal stem cells. Firstly, the invention provides a method for constructing adipose-derived mesenchymal stem cells carrying miR-122. In addition, the invention also discloses an application of the adipose-derived mesenchymal stem cells carrying miR-122 in the preparation of liver cancer chemosensitization preparations. The adipose-derived mesenchymal stem cells (AMSC) carrying miR-122 are used for preparing a chemosensitization agent and are combined with clinically conventional chemotherapeutic drugs so as to be applied to the chemotherapy of a liver cancer tumor-bearing mice model, so that the sensitivity of heterotopic or in-situ tumor tissues to the chemotherapeutic drugs can be obviously improved, the chemotherapy effect is enhanced, and the progress of the liver cancer is blocked. The dosage of the chemotherapeutic drugs can be reduced, and the chemotherapy side reaction is alleviated. The invention provides a technical support for preparing a novel liver cancer chemotherapy targeting sensitizer and a new method for the system chemotherapy of the liver cancer.

Description

technical field [0001] The invention belongs to the field of stem cells and biotechnology, and relates to new applications of mesenchymal stem cells, in particular to a method for constructing adipose-derived mesenchymal stem cells carrying miR-122 and its application in preparing liver cancer chemotherapy sensitization preparations. Background technique [0002] Primary liver cancer (hepatocellular carcinoma, HCC) is one of the common malignant tumors, and its mortality rate ranks third in the global malignant tumor mortality. In my country, the mortality rate of liver cancer is second only to gastric cancer, ranking second in the mortality rate of malignant tumors in my country. According to statistics, about 110,000 people die from liver cancer every year in my country, which is the country with the highest incidence and mortality rate of liver cancer in the world. Chemotherapy is one of the main means of tumor treatment at present, and it has shown positive significance...
