Embodiment Construction

[0015] In order to make the objectives, technical solutions and advantages of the present invention clearer, the present invention will be further described in detail below in conjunction with embodiments. It should be understood that the specific embodiments described herein are only used to explain the present invention, but not to limit the present invention.

[0016] A method for removing PVS virus of potato test-tube plantlets, which is characterized in that it comprises the following steps:

[0017] (1) Pretreatment of test tube seedling materials

[0018] The test tube seedlings containing PVS were tested to contain the top cut sections and placed in a culture flask containing 30-40mg / L ribavirin and 10-20mg / L butyryl hydrazide in MS solid medium, 15 plants per bottle, at a temperature of 23~ 25℃, light time 15h / d, light intensity 2000~3000lx, culture for 15~20d, remove the bottle seedlings contaminated by bacteria and fungi. When the longest root of the new seedling is 0.5cm...

Abstract

The invention provides a method for removing PVS viruses of potato test-tube plantlets. The method comprises the steps of stripping the stem tips of the pretreated test-tube plantlets and then putting the test-tube plantlets into a stem tip culture medium with the stem tip facing upwards and the section facing downwards for culture, next, culturing the stem tips for 40-50 days under the conditions of a temperature within the range of 22+ / -2 DEG C, the illumination time of 12h-15h / d and the illumination intensity of 2000-3000lx, in the period, transferring the stem tips above 0.5cm long to an MS culture medium, and continuously culturing the undifferentiated stem tip callus or small plantlets less than 0.5cm long, and finally, detecting the stem tip seedlings by use of enzyme linked immunosorbent assay, and remaining the satisfied stem tip seedlings, thereby obtaining the potato seedlings without PVS through the whole period of 120-180 days. The method is high in PVS virus removal rate, suitable for batch operations of removing the PVS viruses of the potato variety resources, and capable of obtaining the satisfied seedlings at a time and avoiding repeated labor; besides, the method is high in virus removal rate and stable in virus removal effect, and the detection cost can be reduced.

Description

Technical field [0001] The invention belongs to the technical field of agricultural seedling cultivation, and in particular relates to a method for removing PVS virus of potato test tube plantlets. Background technique [0002] There are more than 5.7 million ha of potato planting in China 2 , Ranking first in the world, but the output level is low (15.45t·hm -2 ), not only lower than the world average yield (17.26t·hm -2 ), not as good as developed countries in the world (50.25t·hm -2 ). Virus disease damage is one of the main factors affecting potato production in China. As potatoes are vegetative reproduction, the virus accumulates year by year as the potatoes multiply, leading to the degradation of potato seed. Studies have shown that potato viruses cause potato production to decrease by 30% to 50%. Mixed infection of different viruses can cause more than 80% of yield loss. According to reports, there are more than 30 viruses that infect potatoes. Potato virus X (PVX) (10-20...
