Problems solved by technology

As far as the extraction process is concerned, the water extraction and alcohol precipitation method takes a long time and the extraction rate is low. Although the alkali extraction method improves the extraction rate to a certain extent, it reduces the product quality. The efficiency has also been greatly improved, but due to limited conditions, it is difficult to use in industrial production

Embodiment Construction

[0026] The present invention will be described in detail below in conjunction with the accompanying drawings.

[0027] 1. Treatment of pumpkin samples and determination of extraction conditions: Wash the pumpkins, peel them, remove their flesh, crush them with a pulverizer, stir them evenly, freeze them at a low temperature for 3 hours, and then use ultrasonic-assisted extraction. The extraction temperature is 60°C, and the ultrasonic power is 200W. The extraction time is 30min, and the ratio of solid to liquid is 1:30.

[0028] 1.1 The effect of freezing temperature on the extraction rate of pumpkin polysaccharides

[0029] The extraction rates of pumpkin polysaccharides under different freezing temperatures are shown in figure 1 . Depend on figure 1 It can be seen that the extraction rate of pumpkin polysaccharides after freezing treatment is much higher than that of pumpkin polysaccharides stored at room temperature (18°C), and as the freezing temperature decreases, the ...

Abstract

The invention discloses an extraction and purification method of pumpkin polysaccharide. The method includes the steps of conducting cleaning, peeling and pulp discarding on pumpkin, smashing the pumpkin through a smashing machine, conducting even stirring and low-temperature freezing, then extracting pumpkin polysaccharide with the aid of ultrasonic waves to obtain a pumpkin polysaccharide extraction solution, adding polyethylene glycol and ammonium sulfate to the pumpkin polysaccharide extraction solution for extraction, conducting sufficient and even stirring, standing till phase separation is completed, conducting centrifugal separation to form two phases of the solution, adding absolute ethyl alcohol to the upper phase, and conducting sedimentation to obtain pumpkin polysaccharide. The method has the advantages of saving time, being efficient, saving energy and the like when ultrasonic waves are used for extracting pumpkin polysaccharide. The inactivation or degeneration of bioactive substances can not be caused when extraction is conducted in a water solution. The yield of pumpkin polysaccharide in the method can reach 60.58-62.82%. The phase separation time is short, and the natural phase separation time is usually 5-15 min. No organic matter residual problem exists, and engineering amplification and continuous operation are easy.

Description

technical field [0001] The invention relates to biotechnology, in particular to pumpkin polysaccharide and its extraction and purification method. Background technique [0002] Pumpkin, also known as pumpkin, pumpkin, pumpkin, etc., is an annual vine herb of Cucurbitaceae, and is one of the important medicinal and edible plants. Pumpkin is rich in nutrition, not only contains mannitol, a variety of amino acids, vitamin C, E and trace elements Fe, Se, Zn, Mn, etc., but also contains a large number of active ingredients such as alkaloids, cucurbitine, and pectin. [0003] Pumpkin polysaccharide is a non-specific immune enhancer, which can improve the immune function of the body, promote the production of cytokines, and play a variety of regulatory functions on the immune system by activating complement and other ways. Pumpkin polysaccharide is brown powder, which is one of the most important and most potential functional factors in pumpkin active ingredients. Not only has co...
